Georgia Mozell, Eve Marks and Maddy Mozell are adult sisters. Georgia (Keaton) is the editor of her own wildly successful self-titled women’s magazine. She strives for publicity at any cost. Party planner Eve (Ryan) is the mother hen of the group, not only of her own family, but also of her siblings and father as their mother, Pat (Leachman), not only emotionally left their father when they divorced, but her daughters as well. And Maddy (Kudrow) is a vacuous soap opera actress who has always struggled for her own identity. Despite being as busy with her own life as the others, Eve is the only one of the three who deals with the long term hospitalization of their cantankerous seventy-nine-year-old father, Lou Mozell, when he enters the early stages of dementia, and the associated outcomes of that hospitalization. Eve’s caring for Lou is despite an especially hurtful incident with him seven years earlier. As the emotional aspect of looking after Lou becomes more and more stressful, Eve has to figure out how to maintain her own sanity, while dealing with her sisters, who believe they too are part of their father’s care while they don’t lift a finger to help.

Sally 'Tally' Atwater (Michelle Pfeiffer) is an ambitious, aspiring news reporter, who is hired by Miami local news director Warren Justice (Robert Redford) when she sends in a homemade audition tape. He carefully guides her career to new heights, all the while becoming increasingly attracted to her. Tally soon rises through the ranks of network news to become successful, while Warren's once-stellar career sinks into mediocrity. Furthermore, Tally's ascension takes her away from Warren when she is forced to relocate to Philadelphia. Tally struggles at her new post, in no small part due to the hostility of veteran reporter Marcia McGrath (Stockard Channing), who jealously protects her position as the top reporter. Warren turns up to inspire Tally, and the two partners begin a new career together. However, on a routine assignment in a Philadelphia prison, Tally and her cameraman are taken hostage in a prison riot and forced to endure hours of intense violence. Tally covers the groundbreaking story from within the walls of the collapsing prison as Warren looks on from outside, guiding her through her first national broadcast. This incredible act of bravery leads to Tally's eagerly anticipated advancement to a national network newscaster position and the continuation of the dynamic duo's rise to fame - but shortly after, disaster strikes when Warren is killed during an assignment.

In 1973, Sam "Ace" Rothstein (Robert De Niro) is a sports handicapper and Mafia associate who is sent to Las Vegas to run the Teamsters Union-funded Tangiers Casino on behalf of the Chicago Outfit. He hires old friend Billy Sherbert (Don Rickles) as his manager. In between, Ace and his friend, mob enforcer and caporegime Nicholas "Nicky" Santoro (Joe Pesci), narrate how the mob bosses control the Teamsters Union, which gives out money for casinos that they own, such as The Tangiers, and how they also drive off rival crews and get rid of cheaters. Ace becomes the Tangiers' de facto boss by taking advantage of lax gaming laws allowing him to work at the casino while his gaming license is still pending. He doubles the casino's profits, which are skimmed by the Mob before the records are reported to income tax agencies. The bosses are impressed with Ace's work and send Nicky to protect Ace and the whole business, along with Nicky's brother Dominick, Nicky's friend and subordinate Frank Marino (Frank Vincent), and the rest of Nicky's soldiers in his crew. Nicky, however, becomes more of a liability than an asset; his criminal activities- which he makes nearly no effort to conceal- and his violent and vicious temper quickly gets him banned by the gaming board from every casino, and his name is placed in the Black Book. In retaliation, Nicky gathers his own crew, opens a jewelry store and restaurant, begins running unsanctioned shakedowns and burglaries, and soon after is considered the mob boss of Vegas.
