In the opening scene, John Hay Forrest (George Gaynes), noted scientist and cheesemaker, dies in a single-vehicle car accident (represented by the car wreck scene from Keeper of the Flame). In the next scene, private investigator Rigby Reardon (Steve Martin) is reading a newspaper when Forrest's daughter, Juliet (Rachel Ward), enters his office and faints when the paper's headline reminds her of her father's death. Upon coming to, she hires Rigby to investigate the death, which she thinks was murder. In Dr. Forrest's lab, Rigby finds two lists, one titled "Friends of Carlotta" and the other "Enemies of Carlotta", as well as an affectionately autographed photo of singer Kitty Collins, whose name appears on one of the lists. His search is interrupted by a man posing as an exterminator (Alan Ladd, in This Gun for Hire), who shoots Rigby in the arm and frisks the lists from the seemingly dead investigator.

In the near future, an atomic disaster has reduced the world to poverty. Instead of a government, America is run by an organization called the Merchants, who exploit the degenerate remains of society. In order to keep control of the populace, the Merchants force Dr. Paul Dean (Robert Glaudini) to create a new life form, a parasite that feeds on its host. Realizing the deadly potential of such a being, Dean escapes the Merchants with the parasite, infecting himself in the process. Now on the run, he travels from town to town, studying the parasite so that he can find a way to destroy it, all the while keeping one step ahead of a Merchant named Wolf (James Davidson) who is hunting for him. While resting in a desert town, he is attacked by a gang of hooligans (Cherie Currie, Freddy Moore, Natalie May, Joanelle Romero, Tom Villard) led by Ricus (Luca Bercovici), a former slave of the Merchants. The gang steal a silver canister containing the parasite, not realizing what it is, and it escapes and infects one of the members. Meanwhile, Paul befriends a pretty young lemon grower named Patricia Welles (Demi Moore), who promises to help him destroy the escaped parasite. Ricus, trying to save the life of his friend, comes to Paul for help, only to be confronted by Wolf. Patricia, Paul, and Ricus manage to evade Wolf, but when they return, the parasite has spread to another member and grown into a fleshy worm with a mouthful of deadly teeth. Ricus becomes a turncoat and attempts to help, but ends up getting killed by Wolf. A friendly diner owner, named Collins, comes to aid the group. After Patricia helps kill the parasite bonded to Paul, which they electrocute, the worm attacks Wolf who is then blown up by Patricia, Paul, and Collins.

In the opening scene, Howard Hughes loses control of and crashes his motorcycle in the Nevada desert. That night, he's discovered lying on the side of a stretch of U.S. Highway 95 where Melvin Dummar stops his pickup truck so he can relieve himself. The disheveled stranger, refusing to allow the Good Samaritan to take him for medical help, asks him to drive him to Las Vegas. En route, the two engage in stilted conversation until Dummar cajoles his passenger into joining him in singing a Christmas song he wrote. Hughes then suggests they sing his favorite song Bye Bye Blackbird, and they do. The man warms up to his rescuer and before he's dropped off at the Desert Inn (which Hughes owns and therein resides), he identifies himself as the reclusive billionaire.

Tommy Chong plays Anthony "Man" Stoner, a jobless, marijuana-smoking drummer who is told to either get a job by sundown or be sent off to military school by his parents. Anthony leaves the house in a 1967 Volkswagen Beetle convertible (which had his father's Rolls Royce radiator grille on the front), a car which is subsequently left smoking on the side of the road. Anthony is picked up while hitchhiking by the equally enthusiastic smoker Pedro de Pacas (Cheech Marin). The license plate reads MUF DVR ("Muff Diver"). They share a large joint, which Man says is made with "mostly Maui wowie" and "Labrador" (essentially dog feces, as his dog, a Labrador Retriever, had eaten his stash). Police find their car parked on a traffic median with them in it, discover that they are clearly stoned and arrest them. At trial, the pair are released on a technicality after the judge's water pitcher is discovered to be containing vodka.

During a space flight to Saturn, three astronauts are exposed to a blast of radiation which kills two of them and seriously injures the third, Steve West (Rebar). He is next shown unconscious in a hospital back on Earth, with bandages covering his face; his physician, Dr. Loring (Lisle Wilson), cannot explain what is happening to West or how he survived the blast. After the doctor leaves, West awakens and is horrified to find the flesh on his face and hands melting away. Hysterical, he attacks and kills a nurse (Bonnie Inch), then escapes the hospital in a panic. Loring and Dr. Theodore "Ted" Nelson (DeBenning), a scientist and friend of West, discover that the nurse's corpse is emitting feeble radiation, and realize West's body has become radioactive. This film picks up the story 15 years after the events of the first film. Drum (Norton) has been born to a white prostitute (Vega), who raises him with her black lesbian lover (Kelly). Drum grows up to be a fighter and is often forced to bare-knuckle-box other slaves for the entertainment of the owners, one of whom is a gay Frenchman named Bernard DeMarigny (Colicos). DeMarigny wants to sleep with Drum, but his advances are rejected by the slave and DeMarigny vows revenge against Drum. Drum and his friend Blaise (Kotto) are eventually sold to plantation owner Hammond Maxwell (Oates) and are both taken to his plantation to work. Drum's girlfriend Regine (Grier) is purchased by Maxwell as well and is taken to the plantation for his own personal desires.
