Le Docteur Knock est un ancien voyou devenu médecin qui débarque dans le petit village de Saint-Maurice afin d'y faire fortune selon une méthode particulière. Suivant son adage « Tout homme bien portant est un malade qui s'ignore. », il va ainsi faire croire aux villageois en les manipulant astucieusement, qu'ils ne sont pas aussi bien portants qu'ils pourraient le penser. C'est ainsi qu'il trouvera chez chacun un symptôme, imaginaire le plus souvent, et de ce fait pourra exercer lucrativement sa profession. Sous ses airs de séducteur et après avoir acquis la confiance du village, Knock est sur le point de parvenir à ses fins. Mais son passé le rattrape et une ancienne connaissance vient perturber les plans du docteur.

Margaret is a beautiful and successful businesswoman selling power plants to the Chinese. With an adoring English lover, she appears to have everything going for her. On her fortieth birthday, Margaret receives the first bundle of letters she wrote to herself when she was seven years old. A jumble of colorful collages, photographs, and wildly creative puzzles seem to have come from a different girl entirely. In a letter the seven-year-old Margaret writes, "Dear me. Today I am seven years old and I'm writing you this letter to help you remember the promises I made when I was 7, and also to remind you of what I want to become..." As her letters to herself keep arriving, Margaret finds herself becoming disenchanted. The letters evoke long-forgotten memories and cast doubt on many of the choices she made in her life. In many ways she's become the opposite of what she hoped to become as a child. Margaret visits her childhood village and, by reconnecting with people who see in her the girl they once knew, she starts finding her way to the woman she vowed to become.

Genetic engineers Clive Nicoli (Adrien Brody) and Elsa Kast (Sarah Polley) hope to achieve fame by splicing animal DNA to create hybrids for medical use at the company N.E.R.D. (short for Nucleic Exchange Research and Development). Their work previously yielded Fred, a dog-sized vermiform creature intended as a mate for their female specimen, Ginger. After successfully mating them, Clive and Elsa plan to create a human-animal hybrid that could revolutionize science. Their employers Joan Chorot (Simona Maicanescu) of N.E.R.D. and William Barlow (David Hewlett) forbid them from doing this. Instead, they are to find and extract proteins used for commercial drug production from Fred and Ginger. Clive and Elsa pursue their own agenda in secret. They develop a viable female creature (Abigail Chu).
