In 1943, the Axis powers plan an assault on the island of Keros — where 2,000 British soldiers are marooned — to bully neutral Turkey into joining them with a display of their military strength. Rescue by the Royal Navy is prevented by two massive radar-directed superguns on the nearby island of Navarone. In Fort Griffin, Texas, Ed Bailey (Lee Van Cleef) comes looking to avenge the death of his brother at the hands of gunslinger John H. "Doc" Holliday (Kirk Douglas). Seeing him in a bar, Holliday's girl, Kate Fisher (Jo Van Fleet), returns to Holliday's room, where the two argue—while Holliday throws knives at the wall—once she brings up Holliday's once-prominent family. At the same time, well-known marshal Wyatt Earp (Burt Lancaster) arrives in Fort Griffin thinking he will take outlaws Ike Clanton (Lyle Bettger) and Johnny Ringo (John Ireland) into custody, but instead finds out that the local sheriff, Cotton Wilson (Frank Faylen), released them despite the outstanding warrants for their arrest. Holliday refuses to help the lawman, holding a grudge against Wyatt's brother, Morgan. Holliday kills Bailey with a knife-throw when Bailey attempts to shoot him in the back. The governor of an unnamed western state, Hubert "Happy" Hopper (Guy Kibbee), has to pick a replacement for recently deceased U.S. Senator Sam Foley. His corrupt political boss, Jim Taylor (Edward Arnold), pressures Hopper to choose his handpicked stooge, while popular committees want a reformer, Henry Hill. The governor's children want him to select Jefferson Smith (James Stewart), the head of the Boy Rangers. Unable to make up his mind between Taylor's stooge and the reformer, Hopper decides to flip a coin. Thomas Dunson (John Wayne) is a stubborn man who wants nothing more than to start up a successful cattle ranch in Texas. Shortly after he begins his journey to Texas with his trail hand, Nadine Groot (Walter Brennan), Dunson learns that his love interest (Coleen Gray), whom he had told to stay behind with the wagon train with the understanding that he would send for her later, was killed in an Indian attack. Despite this tragedy, Dunson and Groot press on. That night, Dunson and Groot, keeping watch, hear a group of Indians planning to attack them. They kill the Indians, and on the wrist of one, Dunson finds a bracelet he had been left by his late mother. One day before, he had presented it to his young love as he left the wagon train. The bracelet reappears significantly later in the film.

In Honolulu, a DC-4 airliner prepares to take off for San Francisco with 17 passengers and a crew of 5. The film is set in Jennings County, Indiana in 1863. In the town of Rio Bravo, Texas, sheriff's deputy Dude (Dean Martin), who has acquired the contemptuous nickname Borrachón ([bo.raˈtʃon], Spanish for "drunk"), enters a saloon but can't afford a drink. Joe Burdette (Claude Akins), brother of rancher Nathan Burdette, tosses a silver dollar into a spittoon. Presidio County, Texas Sheriff John T. Chance (John Wayne) appears and kicks the spittoon away, looking at Dude with disgust. Dude is shamed by his plight and takes out his anger on Chance, knocking him out with an ax handle. In the winter of 180 A.D., the Roman emperor Marcus Aurelius (Guinness) fights to keep Germanic barbarians from invading his northern territories on the Danube frontier. His deputies are the Greek ex-slave Timonides (Mason), a closet Christian, and the stern and honest general Gaius Livius (Boyd). Livius has close connections with the imperial family, being the lover of Aurelius' philosopher daughter Lucilla (Loren) and a friend of her brother Commodus (Plummer). Nevertheless, he is amazed to hear that Aurelius wants to make him his heir. Despite his military obligations the emperor has egalitarian ideals, dreaming of a day when Rome grants equal rights to men of all nations. He knows that he will not live to achieve this end, and trusts Livius to do so more than his charismatic but brutal son. The discovery that his father has effectively disinherited him hurts Commodus immensely, and damages the almost brotherly relationship he had enjoyed with Livius.

The Zacharys are a thriving and respected family on the Texas frontier. Father Will Zachary was killed by Kiowa Indians, leaving his oldest son Ben (Burt Lancaster) as the head of the family.
