In the year 1962, the Radcliffe family and their one hundred and one dalmatians are preparing to move to their "Dalmatian Plantation," a home in the countryside with plenty of room for all of them. However, one of the puppies, Patch, feels ignored and wishes to be unique like his television hero, Thunderbolt. While watching the Thunderbolt Adventure Hour, he hears about a chance to appear on the show. He is accidentally left behind when his family leaves for the plantation, so he decides to head for the audition to meet his hero and win a guest spot on the show. He fails to impress the producers, allowing a jealous Lil' Lightning to manipulate them into making himself the hero of his own show. Depressed, Patch leaves and runs into Thunderbolt.

The film follows 17-year-old Cassandra Mortmain (Romola Garai), and the fortunes of her eccentric family, struggling to survive in genteel poverty in a decaying English castle. The characters include: Cassandra's father (Bill Nighy), a writer who hasn't written anything in the twelve years since the spectacular success of his first novel; her exquisite older sister Rose (Rose Byrne) who rails against their fate and hopes to marry for money; and their bohemian stepmother, Topaz (Tara Fitzgerald), an ex-model still apt to sun-bathe in the nude. The possibility of salvation seems to loom in the form of their wealthy American landlord Simon Cotton (Henry Thomas) and his brother Neil (Marc Blucas). Although Simon initially turns her away, Rose is determined to make him fall in love with her and succeeds. A wedding is arranged and Cassandra appears left on the sidelines. But events spiral out of control, and before the summer ends many expectations will have been overturned.

After three years in prison, Cruella de Vil (Glenn Close) has been cured of her desire for fur coats by Dr. Pavlov (David Horovitch) and is released into the custody of the probation office on the provision that she will be forced to pay the remainder of her fortune (eight million pounds) to all the dog shelters in the borough of Westminster should she repeat her crime. Cruella therefore mends her working relationship with her valet Alonso (Tim McInnerny) and has him lock away all her fur coats. Cruella's probation officer, Chloe Simon (Alice Evans), nevertheless suspects her, partly because Chloe is the owner of the now-adult Dipstick (one of the original 15 puppies of the previous film). Dipstick's mate Dottie has recently given birth to three puppies: Domino, Little Dipper, and Oddball, whereof Oddball lacks spots.

American video game designer Roger Dearly (Jeff Daniels) lives with his pet dalmatian, Pongo, in London. One day, he takes Pongo for a walk in St. James's Park, and he sets his eyes on a beautiful female dalmatian named Perdita. After a frantic chase through the streets of London, Roger and Pongo discover that Perdita likes Pongo; and her owner, Anita Campbell-Green (Joely Richardson), falls in love with Roger when they meet. With his upscale department store steadily losing money, Gabriel Service, Sr. (Lewis Stone) is forced to discharge some of his employees, including an unenterprising but loyal and long-serving Tim Benton (Lionel Barrymore). Then Service returns home to his mansion to inform his family of their own financial straits; he has kept the 200-year-old family firm afloat with his own money in recent times. Neither his young adult children, Caroline and Michael, nor their stepmother Isobel take him very seriously at first, despite his repeated warnings over the past few months.
