The Asgardian Loki encounters the Other, the leader of an extraterrestrial race known as the Chitauri. In exchange for retrieving the Tesseract, a powerful energy source of unknown potential, the Other promises Loki an army with which he can subjugate Earth. Nick Fury, director of the espionage agency S.H.I.E.L.D., and his lieutenant Agent Maria Hill arrive at a remote research facility during an evacuation, where physicist Dr. Erik Selvig is leading a research team experimenting on the Tesseract. Agent Phil Coulson explains that the object has begun radiating an unusual form of energy. The Tesseract suddenly activates and opens a wormhole, allowing Loki to reach Earth. Loki takes the Tesseract and uses his scepter to enslave Selvig and a couple of agents, including Clint Barton, to aid him in his getaway.

Genius, billionaire, and playboy Tony Stark, who has inherited the defense contractor Stark Industries from his father, is in war-torn Afghanistan with his friend and military liaison, Lieutenant Colonel James Rhodes to demonstrate the new "Jericho" missile. The convoy is ambushed and Stark is critically wounded by one of his own rocket propelled grenades. He is captured and imprisoned in a cave by the terrorist group the Ten Rings, and an electromagnet is grafted into his chest by fellow captive Yinsen to keep the shrapnel shell shards that wounded him from reaching his heart and killing him. Ten Rings leader Raza offers Stark freedom in exchange for building a Jericho missile for the group, but Tony and Yinsen agree Raza will not keep his word.

Jim Street (Colin Farrell), a former U.S. Navy SEAL and hot-shot cop from the Los Angeles Police Department and his SWAT team are sent to stop a gang of robbers who have taken over a bank. His high-tempered partner and close friend Brian Gamble (Jeremy Renner) disobeys an order to not fire on the bank robbers, and accidentally wounds a hostage in the process. Gamble and Street are demoted by Captain Fuller (Larry Poindexter), the commander of the LAPD Metropolitan Division. Gamble quits following the argument with Fuller, and Street is taken off the team and sent to work in the "gun cage", where he looks after the gear and weaponry. Six months after the incident, the chief of police calls on Sergeant Daniel "Hondo" Harrelson (Samuel L. Jackson) to help re-organize the SWAT division. In the coastal town of Astoria, Oregon approximately six months after the events of the first movie, a teenage boy named Jake asks a classmate named Emily over to his house, under the guise of studying together; however, his motives are actually centered on having her watch Samara Morgan's cursed video tape, as his seven-day deadline is fast approaching. Emily plays the tape while Jake waits in the kitchen, but when he returns to the living room, it is revealed that she covered her eyes throughout the tape, and thus didn't see any of it. As she failed to perpetuate the guidelines of the tape's curse, Samara crawls out of the television and kills Jake in front of Emily.

Over the two years since the death of Deacon Frost, Blade (Wesley Snipes), the Daywalker, a vampire unaffected by sunlight and who chooses to defend humans and fight other vampires, has been trying to find his mentor Whistler (Kris Kristofferson), who survived his suicide attempt in the first crisis. Blade finds Whistler locked in a tank by a gang of vampires who were keeping him alive to torture. Blade rescues Whistler and brings him back to the lair, ridding the latter's vampirism with a cure developed by Dr. Karen Jenson.

In 1898, Sir Robert Beaumont (Tom Wilkinson), the primary financier of a railroad project in Tsavo, Kenya, is furious because the project is running behind schedule. He seeks out the expertise of John Henry Patterson (Val Kilmer), an Irish military engineer, to get the project back on track. Patterson travels from England to Tsavo, telling his wife, Helena, he will complete the project and be back in London for the birth of their son.
