In Mexico, a freelance vampire slayer, Derek Bliss (Jon Bon Jovi), is hired by an unknown client provided he build a team of slayers. Father Adam Guiteau (played by Tim Guinee in the first film) is shown to have been killed in this one. This team ends up including the vampiress Zoe, who is fighting her affliction with medication, Father Rodrigo, a helper who pretends to be a priest, and Sancho, a brave fourteen-year-old boy who aids Derek in killing some vampires at the beginning of the movie, in finding a large nest of "suckers" and their powerful leader, a vampire princess named Una (Arly Jover). She is seeking a legendary black crucifix: the Berziers Cross, the same crucifix used unsuccessfully in the first movie to perform a ritual which will enable vampires to walk in sunlight and be invulnerable.

On the streets of Fifth Avenue, an orphaned kitten named Oliver is left abandoned after his fellow orphaned kittens are adopted by passersby. Wandering the streets by himself in search of someone to adopt him, Oliver meets a laid-back dog named Dodger who assists the kitten in stealing food from a hot dog vendor named Louie. Dodger then flees the scene without sharing his bounty with Oliver. Oliver follows Dodger all throughout the streets until he eventually arrives at the barge of his owner, a pickpocket named Fagin, along with his meal, to give to his friends: Tito the Chihuahua, Einstein the Great Dane, Rita the Saluki and Francis the Bulldog. Oliver sneaks into their home, located below the city's docks, and is discovered by the dogs. After a moment of confusion, Oliver is then received with a warm welcome. Fagin, owner of the dogs, comes in and explains that he is running out of time to repay the money he borrowed from Sykes, a ruthless shipyard agent and loan shark. Sykes tells Fagin the money must be paid in three days, or else. Sykes' Dobermans Roscoe and DeSoto attack Oliver but the cat is defended by Fagin's dogs. Immediately thereafter, a depressed Fagin returns to the barge, lamenting that he has only three days to find the money he owes Sykes. After the dogs cheer him up, Fagin is introduced to Oliver, and, considering that they all need help, accepts him into the gang.

Situated in the early 1980s, the plot focuses on Cindy Fralick, a parking enforcement officer who is uninspired by her job. Jesse Lujack (Richard Gere) is a cocky, nihilistic drifter in Las Vegas, obsessed with Marvel Silver Surfer comic books, the rock and roll music of Jerry Lee Lewis and Monica Poiccard (Valérie Kaprisky), a UCLA architecture undergraduate whom he knows only from a weekend fling in Vegas. As the film opens late at night, he steals a Porsche, intending to drive to Los Angeles. As he speeds down the highway and looks through the owner's possessions, he discovers a handgun in the vehicle's glovebox. Seeing his reckless driving, a policeman gives chase and Jesse is forced off the road and becomes stuck. When the policeman orders him to step away from the car, Lujack impulsively grabs the gun and (apparently accidentally) shoots the officer. Fleeing to Los Angeles, Lujack finds his picture splashed all over the newspaper and TV news as the "cop killer.
