The film begins in the Elizabethan era shortly before the death of Queen Elizabeth I. On her deathbed, Elizabeth promises an androgynous young nobleman named Orlando a large tract of land and a castle built on it along with a generous monetary gift which she will only bequeath to him if he consents to her command, "Do not fade. Do not wither. Do not grow old." Both he and his heirs will keep the land and inheritance forever. Orlando acquiesces and resides in splendid isolation in the castle for a couple of centuries, during which time he dabbles in poetry and art. His attempts to befriend a celebrated poet, however, backfire when the poet ridicules his verse. Orlando then travels to Constantinople as British ambassador to the Turks, but he is almost killed in a diplomatic fracas there. Waking up the next morning, he learns something even more startling: he has physically transformed into a woman overnight.

Ralph Milan is a contract killer who is paid to kill Louis Randoni, whose testimony in various trials could harm the organisation. Ralph waits for his prey in his hotel room, but is interrupted by his comical neighbour, a shirt salesman named François Pignon (Jacques Brel). Pignon, who is suicidal since his wife left him for a reputed doctor named Fuchs (Jean-Pierre Darras), tries to hang himself on the waterpipes, but only manages to cause a flood. Realizing that if Pignon tries to kill himself again the police will search the place, Milan offers to talk him out of it until after his assassination. Unfortunately Pignon starts irritating him more, and makes it more difficult for him to fulfill his contract killing.

François 22 ans, travaille dans une banque au milieu d’une agitation permanente qui lui paraît stérile. Il est très mélancolique et semble ne plus avoir le goût à la vie, il passe le plus clair de son temps à se droguer et à s'isoler dans sa chambre, ne s'alimentant presque plus, causant le désespoir de ses parents chez qui François vit et aussi celui de sa copine Sophie, une danseuse à l'Opéra. Quand il ne se drogue pas, François voit ses amis, Guy qu'il connait de longue date et Pierrot, un garçon un peu plus jeune. Multipliant les absences à son travail, François finit par en être renvoyé. La tristesse et la déception de la vie de François s'intensifient peu à peu. Il finit notamment par se séparer de Sophie. Alors qu'il décide de quitter Paris, il meurt d'une overdose. Sa mère, ne supportant pas sa mort se suicide quelques jours plus tard. Sur leur tombe commune, se recueille Guy qui dit une phrase qu'avait l'habitude d'écrire son ami : "Je croyais que la vie était un poème".
