"Lila" ‒ played by Susan Stewart ‒ is a seemingly good-natured go-go dancer who strips at a seedy topless bar on the Sunset Strip. After taking LSD, Lila becomes a psychopathic serial killer. She continues to pick up men at the bar where she is employed, but after her sanity is lost she routinely is interrupted mid-coitus by psychedelic bad trips in which she visualizes a balding, half-naked old man clutching wads of cash in one hand and a bunch of bananas in the other. These psychotic episodes cause her to murder her partners by stabbing them with a screwdriver and dismembering them with a rusty meat cleaver (or in one case, a garden hoe) while imagining that she is cutting up cantaloupes and watermelons.

The film presents a snapshot of '60s San Francisco before shifting its focus to strippers. The strippers' lives are earnestly portrayed as they reveal the day-to-day realities of sex work, talk bra sizes, relate their preferences in men, all voiced over while dancing topless to a 60s instrumental rock soundtrack. Throughout a large portion of the film, the narrator talks about the women as if they are a sub-genre of the counter culture movement, somewhat similar to the beatnik or hippie movements that were highly prevalent during the same era. The "Topless" movement as it is called by the narrator could also be perceived as an allegorical subset of the Sexual Revolution of the 1960s.

The film opens to two muscle-bound men dressed in loincloths approaching a crypt. They open the doors, revealing a coffin. They remove the lid and exit the crypt, then the inhabitant of the coffin (Criswell) sits up to deliver an opening narration. This narration mostly matches the prologue of Night of the Ghouls (1959), with one minor variation and an additional line. The phrase "world between the living and the dead" of the original is changed to "void between...". There is also a new line at the end: "A night with the ghouls, the ghouls reborn, from the innermost depths of the world!" The opening credits feature the image of "an immobile young woman clad in gold". The image was probably inspired by a memorable scene of Goldfinger (1964).
