After the mysterious failure of the Discovery One mission to Jupiter in 2001, which resulted in the deaths of four astronauts and the disappearance of David Bowman, the fiasco was blamed on Dr. Heywood Floyd, who resigned his position as head of the National Council for Astronautics. While an international dispute causes tensions between the United States and the Soviet Union, both nations prepare space missions to determine what happened to the Discovery. Although the Soviet ship, the Leonov, will be ready before the American spacecraft Discovery Two, the Soviets need American astronauts to help board the Discovery and investigate the malfunction of the ship's sentient computer, HAL 9000, which caused the disaster. The US government agrees to a joint mission when it is determined that Discovery will crash into Jupiter's moon Io before Discovery Two is ready. Floyd, along with Discovery designer Walter Curnow and HAL 9000's creator Dr. Chandra, joins the Soviet mission.

In the fall of 2008, amidst both the American financial crisis and the presidential election campaign, a man named Johnny "Squirrel" Amato (Vincent Curatola) plans to rob an illegitimate poker game. He enlists Frankie (Scoot McNairy), a former business associate, and Russell (Ben Mendelsohn), a heroin addicted Australian expatriate who is stealing purebred dogs for money, to perform the robbery. Markie Trattman (Ray Liotta), the proprietor of a poker ring, is revealed to have previously orchestrated an inside job by paying two men to rob his own illegal poker room. He holds up under rough questioning by the hitman Dillon (Sam Shepard), though later he openly admits his involvement to various criminal figures and suffers no retaliation. Squirrel anticipates that the Mafia will automatically blame Markie for the heist.

Set in 1934 Paris, the film opens with Richard Di Nardo, a young hustler, emerging from the bed of middle-aged Carroll Todd (Robert Preston), aka Toddy; Richard dresses, takes money from Toddy's wallet and leaves Toddy's apartment. Going about his day, Toddy, a performer at Chez Lui in Paris, sees Labisse, the club owner, auditioning a frail, impoverished soprano, Victoria Grant (Julie Andrews). After the audition, Labisse drily writes her off, and she responds by sustaining a pitch to shatter his wine glass using resonant frequency. That night, Richard comes to Chez Lui as part of a straight foursome and Toddy incites a brawl by insulting Richard and the women in his group. Labisse fires Toddy and bans him from the club. Walking home, he spots Victoria dining at a restaurant, and she invites him to join her. As neither of them can pay for the meal, she dumps a cockroach in her salad to avoid paying their check, but it escapes and the whole place breaks out in havoc.

London Hospital surgeon Frederick Treves finds John Merrick in a Victorian freak show in London's East End, where he is kept by Mr. Bytes. His head is always hooded, and his "owner," who views him as retarded, is paid by Treves to bring him to the hospital for exams. He shows Merrick to his colleagues and highlights his monstrous skull, which forces him to sleep with his head on his knees, since if he were to lie down, he would asphyxiate. On Merrick’s return he is beaten so hard by Bytes that an apprentice calls Treves to bring him back to hospital. When Bytes accuses Treves of likewise exploiting Merrick for his own ends, he vows to do what he can to help Merrick.

Young, starstruck Robert Ford (Casey Affleck) seeks out Jesse James (Brad Pitt) when the James gang is planning a train robbery in Blue Cut, Missouri, making unsuccessful attempts to join the gang with the help of his older brother Charley (Sam Rockwell), already a member. Lovers Lula and Sailor are separated after he is jailed for killing a man who attacked him with a knife; the assailant, Bobby Ray Lemon, was hired by Lula's mother, Marietta Fortune. Later, back in the room, after making love again, Sailor and Lula finally decide to run away to California, breaking Sailor's parole. Unbeknownst to Farragut, however, Marietta also hires gangster Marcellus Santos to track them, and kill Sailor. Santos's minions capture and kill Farragut, sending Marietta into a guilt-fueled psychosis.

Jeffrey Beaumont (Kyle MacLachlan) returns to his logging home town of Lumberton, North Carolina from Oak Lake College after his father suffers a near-fatal stroke. Alvin Straight has not shown up to his regular bar meeting with his friends. He is eventually found lying on his floor at home, although he insists that he "just needs a bit of help getting up". Jeffrey Beaumont (Kyle MacLachlan) returns to his logging home town of Lumberton, North Carolina from Oak Lake College after his father suffers a near-fatal stroke. While walking home from the hospital, he cuts through a vacant lot and discovers a severed ear. Jeffrey takes the ear to police detective John Williams (George Dickerson), through whom he reacquaints with the detective's daughter, Sandy (Laura Dern). She tells him details about the ear case and a suspicious woman, Dorothy Vallens (Isabella Rossellini), who may be connected to the case. Increasingly curious, Jeffrey enters Dorothy's apartment by posing as an exterminator, and while Dorothy is distracted by a man dressed in a yellow suit at her door (whom Jeffrey later refers to as the Yellow Man), Jeffrey steals her spare key.

Alvin Straight has not shown up to his regular bar meeting with his friends. He is eventually found lying on his floor at home, although he insists that he "just needs a bit of help getting up". His daughter Rose takes her reluctant father to see a doctor, who sternly admonishes Alvin to give up tobacco. He also tells Alvin that he should start using a walker. Alvin refuses, and does not tell Rose. Alvin then learns that his brother Lyle has suffered a stroke. Longing to visit him, but unable to drive, Alvin gradually develops a plan to travel to Mount Zion on his "ancient" riding lawn-mower and towing a small homemade travel-trailer, to the consternation of his family and friends.

In 1921, Polish Catholic sisters Ewa (Marion Cotillard) and Magda (Angela Sarafyan) arrive at Ellis Island, New York City as immigrants looking for a better life after escaping their ravaged home in post–Great War Poland. Magda is quarantined because of her lung disease. Ewa is almost deported, but Bruno (Joaquin Phoenix) notices her and her fluency in English, bribes an officer to let her go, and takes her to his house. Knowing Ewa has to make money to get Magda released, Bruno lets her dance at the Bandits' Roost theater and prostitutes her. Bruno also becomes interested in her romantically.
