Hercules (Dwayne Johnson) is the leader of a band of mercenaries comprising the spear-wielding prophet Amphiaraus (Ian McShane) of Argos, the knife-throwing thief Autolycus (Rufus Sewell) of Sparta, the feral warrior Tydeus (Aksel Hennie) of Thebes, the Amazon archer Atalanta (Ingrid Bolsø Berdal) of Scythia and his nephew storyteller Iolaus (Reece Ritchie) of Athens. Hercules is said to be the demigod son of Zeus, who completed the legendary Twelve Labors, only to be betrayed by Hera who drove him insane and caused him to murder his wife Megara (Irina Shayk) and their children during a visit to King Eurystheus (Joseph Fiennes). Throughout the film, it is not clearly established that Hercules is truly the son of Zeus, and many are skeptical of the claim as well as of the stories of Hercules' famous Twelve Labors. Despite this, Hercules is frequently seen displaying unusual strength and nigh-unmatched skill in combat. Hercules is frequently haunted by the memory of the deaths of his wife and daughters by his hand, as well as visions of Cerberus.

In the royal city of Nasaf, the king Sharaman rules the nation of Persia with the help of his brother Nizam. The king goes on to have two sons named Tus and Garsiv. One day in the marketplace, an orphan boy named Dastan protects his friend, Bis, from being executed by Persian soldiers. The king sees Dastan's bravery and adopts him, making him a prince of Persia. Years later, Dastan is a grown man and is still up to his rebellious ways. However, he remains faithful to the empire. When Nizam receives proof that the city of Alamut is supplying weapons to Persian enemies, he and Tus plan to attack with Garsiv leading the siege. Ignoring Tus’ command, Dastan and Bis breech Alamut on their own, and destroy most of the city. Meanwhile, the princess of Alamut, Tamina, orders her servant Asoka to take a special dagger out of the city and protect it at all costs. During the battle, Dastan and Asoka cross paths and fight; Dastan ultimately wins. He then finds the dagger and keeps it.

Set in 1988, Mark Walsh (Colin Farrell) is a photojournalist who has earned a reputation for working in some of the most unforgiving locations on Earth. When his editor Amy (Juliet Stevenson) asks him to cover Saddam Hussein's campaign against the Kurds, Mark takes the assignment and thinks little of it. In the timeframe of 10,000 BC, a tribe of hunter-gatherers called the Yagahl live in a remote mountain range in the Urals and survive by killing woolly mammoths, which they call "mannaks". The camp is led by a hunter who has proven his bravery by killing a Mammoth, and taking the White Spear. The people also strongly venerate an elderly woman, called Old Mother. Because of her different appearance to other humans in the village, it is assumed she is a Neanderthal, the "last of her kind", living with the Homo sapiens of the camp.
