Prison-A-Go-Go! tells the story of a sweet, innocent young woman named Callista who is abducted for "scientific reasons” and taken far away. Now it is up to her bright sister, Janie to solve the mystery and take a crack at rescuing her little sister. But the only clue she finds leads to a notorious prison in the Philippines, where committing a crime may be the only way in. The film was marketed as being filled with multiple shower scenes, girls running amok, kung fu food fights, ninjas, mutant zombies, evil scientists, genetic mutation, and mud wrestling.

Michael Overton (Michael Esposito) and his wife Allison (Marie Caldare) are a couple who have been traumatized by the death of their daughter in a car accident. Allison has been especially affected, as she has been unable to even acknowledge that her daughter has died. She lives in a trance-like state and is unable to perform normal adult functions. Dr. Turner (John Hurt) recommends the Overtons hire a live-in nurse to assist with Allison's psychological healing. They hire a nurse recommended by Dr. Turner, Katherine Randolph (Beth Tegarden), whose unorthodox methods cause tensions to arise in the Overton home.

Planet Spaceball, led by the incompetent President Skroob, has wasted all of its air. Skroob schemes to steal air from the planet Druidia by kidnapping Princess Vespa, the daughter of King Roland on the day of her pre-arranged wedding to the narcoleptic Prince Valium. Skroob sends the villainous Dark Helmet to complete this task with Spaceball One, an impossibly huge ship helmed by Colonel Sandurz. Before they can arrive, Vespa herself abandons her marriage and flees the planet in her personal Mercedes spaceship along with her Droid of Honor, Dot Matrix.

The film follows Melinda, a female employee of the Pentagon's public information service, who loses her job when she refuses to give in to her male superiors' sexual advances. She then becomes outraged by the Pentagon's "un-patriotic" actions, and takes revenge on them by using her physical charms to seduce the top brass into letting her back into the Pentagon. After being re-employed, she sets out to manipulate them to her will and destroy their careers, thus ridding the US government of what she sees as "perverts", who care more about cheap thrills than national security. She eventually seduces the Russian ambassador into betraying valuable military secrets by offering him among other things "the most scrumptious little breasts" as well as something (left unsaid) which she declares to be the "softest, moistest, sweetest". In the end she is appointed Secretary of Defense as a reward.

The story centers around Isaac Hendrix (portrayed by Turman), a young college student studying law and a taxi-cab driver in New Orleans. While out on a night of fun with his friends and wife, Christella during a hypnosis act, he becomes an unwilling host for the restless spirit of J.D Walker, a hustler killed during the 40s. Over the course of the film "Ike" finds his own personality gradually being taken over by the sociopathic Walker, even eventually going so far as to adopt his hair and fashion style, mannerisms, and psychotic tendencies (including an attempted rape on his wife after she mocked his J.D. haircut). With the spirit of J.D. in complete control he turns his attention toward wreaking vengeance against the man responsible for killing his sister, Theotis Bliss. Ike commits havoc all over town along the way before making his way to the church where Theotis' brother works as a preacher, where he finally reveals himself and instructs Elijah to tell Theotis to meet him "on the killin' floor". Ike's wife has meanwhile gone to her ex-husband, a cop who is out for Ike's blood believing him to be a simple psycho hiding behind a false persona--until he mentions to the Chief that Ike claimed his name was J.D. Walker, a man who was not only real, but had died over 30 years ago. J.D. was a hustler who ran numbers during World War II as well as a black-market meat plant where he was murdered by Theotis Bliss after witnessing the murder of his own sister, Betty Jo, at his hands because of her derisive chiding of him and threatening to expose the secret she held about her baby daughter. After being discovered over Betty Jo's lifeless body with her blood on his hand, Elijah Bliss (Gossett Jr.), Betty Jo's husband and the believed father of her child (and younger, submissive brother of Theotis), accused J.D of being the killer and J.D was gunned down on the spot by Theotis to cover up the event. Following Theotis to the old factory, Elijah finally learns the truth before getting into a struggle with Theotis for his gun, during which the weapon discharges and kills Theotis while Ike watches and laughs maniacally as the event plays out. His business complete, J.D. appears to leave Ike's body and thanks to Elijah's testimony, he is allowed to go free to rejoin his wife and friends waiting for him outside.
