Ex-Major Frank McCloud (Humphrey Bogart) arrives at the Hotel Largo in Key Largo, Florida, to visit the family of George Temple, a friend from the Army who had served under him and was killed in the Italian campaign. He meets with George's widow Nora Temple (Lauren Bacall) and his father James (Lionel Barrymore), who owns the hotel. Because the winter vacation season has ended and a major hurricane is approaching, the hotel has only six guests: the dapper Toots (Harry Lewis), the boorish Curly (Thomas Gomez), stone-faced Ralph (William Haade), servant Angel (Dan Seymour), an attractive woman, Gaye Dawn (Claire Trevor) who suffers from alcoholism, and a sixth man who remains secluded in his room. Elvira Kent (Janis Paige) and her husband Michael (Don DeFore) suspect each other of cheating. For their wedding anniversary, Elvira books an ocean cruise to Rio de Janeiro but her husband claims that unexpected business will prevent him from going. Seeing an opportunity, Elvira pretends to take the trip alone, but in fact sends singer Georgia Garrett (Doris Day), a woman she'd met at the travel agency, in her place and under her name. By secretly staying behind, Elvira hopes to find out if Michael is indeed sneaking around behind her back. Michael, however, is suspicious over Elvira's supposed willingness to go on the trip alone, and so hires private detective Peter Virgil (Jack Carson) to see if she is sneaking around behind his back.

In 1940, German-born engineer Kurt Muller (Paul Lukas), his American wife Sara (Bette Davis), and their children Joshua (Donald Buka), Babette (Janis Wilson), and Bodo (Eric Roberts) cross the Mexican border into the United States to visit Sara's brother David Farrelly (Donald Woods) and their mother Fanny (Lucile Watson) in Washington, D.C. For the past seventeen years, the Muller family has lived in Europe, where Kurt responded to the rise of Nazism by engaging in anti-Fascist activities. Sara tells her family they are seeking peaceful sanctuary on American soil, but their quest is threatened by the presence of houseguest Teck de Brancovis (George Coulouris), an opportunistic Romanian count who has been conspiring with the Germans in the nation's capital.
