Bickford Waner (Hopper) who has failed as a train robber, decides to go straight and get an honest job. He arrives in Dime Box, Texas to find work. He's befriended by Reese Ford (Oates) and his wife Molly (Lee Purcell). Molly seduces Bickford getting herself pregnant and making her husband miserable, feeling betrayed by his best friend. Mean John, (Ben Johnson) the sheriff is right on top of the whole plot, recognizing Kid Blue for what he really is, in spite of his trying to change his character. Another friend that Kid Blue has is the town's preacher Bob, (Peter Boyle) who does an excellent acting job.

Kansas (Hopper) is a stunt coordinator in charge of horses on a western being shot in a small Peruvian village. Following a tragic incident on the set where an actor is killed in a stunt, Kansas decides to quit the movie business and stay in Peru with a local woman. Kansas thinks he has found paradise, but is soon called in to help in a bizarre incident: the Peruvian natives are "filming" their own movie with "cameras" made of sticks, and acting out real western movie violence, as they don't understand movie fakery. The film touches on the ideas of fiction versus reality, especially in regards to cinema. The movie is presented in a way that challenges the viewer's traditional cinematic understanding of storytelling, by presenting the story in a non-chronological fashion, and by including several devices typically only seen behind the scenes of filmmaking (rough edits and "scene missing" cards), and the use of jarring jump cuts.

In New York City, soft-spoken Helen returns to the apartment she shares with her boyfriend Marco after enduring an unhygienic and inept abortion. Although Helen becomes ill from the procedure, Marco is satisfied because he was able to procure the abortion through bartering, and thus did not have to expend any money. When Marco leaves briefly to collect some cash, Bobby, an amiable small-time drug dealer to whom Marco owes money, shows unexpected gentleness and concern for Helen. Soon after, when Helen is hospitalized for profuse bleeding, Bobby sneaks in after visiting hours and tells her that Marco has gone away. Briefly, Helen considers returning to her dysfunctional family in Fort Wayne, Indiana. However, after she is released from the hospital, she moves in with Bobby. As they are walking together, Bobby steals a television out of a parked van and pawns it for money to buy drugs. At his apartment, Bobby declines Helen's invitation to have sex until she feels better. When she awakens to find him taking drugs, he explains that he is not an addict, but only "chipping.

The protagonists are two freewheeling hippies: Wyatt (Fonda), nicknamed "Captain America", and Billy (Hopper). Fonda and Hopper said that these characters' names refer to Wyatt Earp and Billy the Kid. Wyatt dresses in American flag-adorned leather (with an Office of the Secretary of Defense Identification Badge affixed to it), while Billy dresses in Native American-style buckskin pants and shirts and a bushman hat. The former is appreciative of help and of others, while the latter is often hostile and leery of outsiders.

With the rise of Hitler, Sol Nazerman (Steiger), a German-Jewish university professor, was dragged to a concentration camp along with his family. He saw his two children die (one while riding in a cattle car) and his wife raped by Nazi officers in the camp. Now he operates a pawnshop in East Harlem, while living in an anonymous Long Island apartment. Numbed by his experiences, he has worked hard not to experience emotions. Nazerman is bitter and alienated, viewing the people around him as "rejects, scum." He is shown interacting cynically as he bargains with the many desperate characters pawning their goods.
