In the ancient Kingdom of Mahishmati in India, Sivagami, the queen mother (Ramya Krishnan), carrying a baby in her arms, emerges from a cave adjoining a big waterfall. She kills the soldiers pursuing her and sacrifices herself to save the baby. Local villagers spot the queen's hand carrying baby in the river and rescues the infant. Sanga (Rohini) and her husband name the infant Shivudu and raise him as their own son. They seal the cave fearing the child's safety.

Irendri (Lakshmi Manchu) is a sorceress, who captures sarpa sakthi (serpent power) and tries to terrorise people of Anga Rashtram. So, a guru arrests her and doesn't allow her to play with the lives of people and destroys her. Even before she was destroyed with the sarpa sakti, Irendri takes her soul away and traps in a locket. Though she dies, her soul (atma) lives in the locket. Her great great grand daughter is Priya (Shruthi Haasan) and she lives as a gypsy.

Jaidev (Pawan Kalyan) is an accomplished hitman and the most trusted aide of Bhagawan (Jackie Shroff), who is the supreme Don of Kolkata. Jaidev remains loyal to him as he helped in seeking revenge with the people who raped his mother and sister when he came to Kolkata in his childhood after his father's death. Faction feud in Rayalseema results in the death of Ramineedu's (V. Nagineedu) brother and he, along with his two sons Mallasuri (Supreeth Reddy) and Baireddy (Venugopal), vow for revenge. Time turns 28 years and comes to Hyderabad. Here lives Ramu (Sunil), an innocent and somewhat unlucky guy whose parents are no more and is ousted out of his job. His life takes a turn with a correspondence that he has got five acres of land in his hometown in Andhra Pradesh. He decides to sell that land and sets off to the village. In this process, he also meets Aparna (Saloni Aswani) who is the daughter of Ramineedu. However, much to Ramu's bad luck, he happens to be the son of the man who killed Ramineedu's brother and he ends up in their home itself. He takes advantage of a tradition of their house that not even one drop of blood should fall inside the house. Whether he comes alive out of their hands or not forms the rest of the story.

Vedam revolves around 5 principal characters. Vivek Chakravarthy (Manoj Manchu) is an upcoming rock star in Bengaluru who hails from a family of decorated army officers. His mother wants him to follow the same path but he isn't interested. He is about to give his first live concert on New Year's Eve with his band-mates in Hyderabad. However, Vivek is late to catch the flight and so the band is forced to travel by road. After sometime, one of their tyres get deflated. Not having a spare, the other friends go to get the tyre inflated leaving Vivek and Lasya (Lekha Washington) alone. Meanwhile, they are attacked by a Hindu racist group. However, they manage to get out of the mess with the help of a Sikh truck driver (who incidentally gets involved in an earlier altercation with Vivek). The band successfully reaches Hyderabad. However they suffer a minor accident there which involves a mother in labour. So Vivek and Lasya take her to a hospital nearby.

Abhiram (Raghu) is a rich spoilt brat who believes only in class and hates the company of poor people with low profiles. He is in deep love with Dr. Janaki, an orphan who aims to serve poor people as a doctor and give them proper medication. Though he is in love with her, Abhiram does not like the path traversed by Janaki in helping poor and needy. This breaks their relationship apart and Janaki walks away to remote place on her job. Not able to withstand the agony of separation, Abhiram sets on a journey in finding Janaki again to carry on the relationship. Meanwhile, he meets Gaali Seenu (Kitty), a petty thief who is an expert in robbing the two wheeler vehicles. He strikes a deal with Seenu and further embarks his journey in search of Janaki. En route his search, Abhiram begins to slowly realize the real world and the noble intentions Janaki had in her service. Finally, the lovers meet up and what happens next forms the rest of the story.
