The movie starts with a TV crew interviewing Prof. Tiwari (Pankaj Kapur) about the events that had occurred in reference to the movie shootouts and eradication of organized crime in Lucknow and surrounding areas. Since a young age, Ajay Kumar (Arshad Warsi) has been traumatized by the death of his army officer father, who killed himself with his service revolver when accused of being a deserter, leaving his widow Prabha (Suhasini Mulay) to bring up Ajay on her own. Ajay grows up, studies hard and successfully becomes an Indian Police Service officer with the title of Senior Superintendent of Police of Lucknow. Due to his honesty, he gets transferred to various states in India, around 14 times in eight years. At his new posting in Uttar Pradesh, he comes to terms with a new criminal boss and ruthless killer, Gajraj Singh (Sushant Singh). Ajay engineers the setting up of a Special Task Force to deal with Gajraj. However, he also runs into problems as Gajraj is politically well-connected. Gajraj continues to elude the Task Force, using more modern technology such as the use of cell phones. But the Task Force recovers and is able to monitor Gajraj's cell phones with the help of Prof. Tiwari. They find out that Gajraj is planning to contest the elections and if he wins, no police officer of any rank, even the Task Force, would then dare to arrest him or even consider him a suspect, which may well result in the Task Force being mere paper tigers. They have a final confrontation inside a train which leaves from Hazrat Nizamuddin railway station. All the officers in the Task Force as well as members of Gajraj's gang kill each other in the fight, leaving Prof. Tiwari as the only witness.

Small-time crook, Champak Chaudhary alias 'Champ' (Arshad Warsi) forges passports, photographs, and deeds in Thailand. He is arrested by the police, tried and sentenced to six months in jail. This is where he meets a supposedly mute and deaf inmate, Raghu Sharma (Raghuvir Yadav), serving a life sentence for stealing diamonds. Champ finds out that Raghu is not deaf nor mute and offers him a share if he springs him out of prison; Champ agrees. As soon as Champ's term is over, he gets discharged and prepares forged release documents for Raghu and gets him 'legally' out of jail.

Champak Chaturvedi (Paresh Rawal) the director of a theatre group, is offered a chance to perform a show in London. Bunty (Akshay Kumar) and Babla (Govinda) are two derps and flirts in his group, always fighting with each other to get the role of the hero. Their behaviour causes the actress (Tanushree Dutta) to run away. This really upsets the organizer, who offered the chance mainly because of the heroine. After reaching London, Champak tells the duo that whoever gets a new actress will get to become hero. Then Babla asks Gullu (Rajpal Yadav), a local driver where he can find a heroine. Gullu misinterprets this as prostitute and directs him to a park.

Murli Prasad Sharma alias Munna Bhai (Sanjay Dutt), a gangster who is in love with the voice of Janhavi (Vidya Balan), a radio jockey, devises a plan to meet her when she announces a quiz contest featuring the life and beliefs of Mahatma Gandhi, to be aired on 2 October, the birthday of Gandhi. To prepare for the contest, Munna's sidekick Circuit (Arshad Warsi) kidnaps and bribes a group of professors to provide the answers for Munna. After winning the contest, Munna is granted an interview with Janhavi wherein he presents himself as a professor of history and a Gandhi specialist. Janhavi subsequently asks Munna to present a lecture on Gandhi to a community of senior citizens who live in her home, called the 'Second Innings House.' Software engineer Balajeet "Bala" Roy (Sanjay Dutt), is happily married to Nisha Roy (Celina Jaitly), with whom he is having a baby. Dhamaal is about four friends; Boman Contractor (Aashish Chaudhary), who lives a wealthy lifestyle with his eccentric dad, Nari (Asrani), who loves his antique car more than his son, and asks his son to leave when he damages the car; dim-witted Manav (Jaaved Jaffrey), his elder brother Adi (Arshad Warsi); and Roy (Ritesh Deshmukh), a trickster who wears a magnetized belt buckle. The group race towards Goa in Nari's stolen car, little knowing that their plans will soon be foiled, albeit hilariously, by Police Inspector Kabir Nayak (Sanjay Dutt), who wants the money at any cost.
