It is written and directed by Jay Anania, stars Academy Award winner Olympia Dukakis, Carmen Chaplin, Alyssa Sutherland and Martin Donovan and is produced by William Fisch and Larry Rattner. The film was scored by John Medeski with vocals by Judy Kuhn.
Day on Fire was screened at the 2006 Cannes Film Festival on May 22, 2006.
Day on Fire tells the story of a singer, a model, an Arab woman journalist, and a physician as they criss-cross New York City over a 12-hour period. Their intersecting lives unfold against the backdrop of a ghastly suicide bombing in Israel, and the strange New York City wanderings of a malevolent Handsome Man, whose predatory intents lend an air of inevitable, horrific violence. Beautifully and hauntingly musical, this thriller also has a political and personal intrigue that mounts inexorably as the sun begins to set on this fateful day. By the time night has fallen in the city, the crossing of these individual fates is sealed. Each of these four women has found their destiny amidst the brutality that the film reveals.

Mollie is an accountant living in New York City who has an affair with Albert, a womanizing executive who is married with two children, and becomes pregnant. During her pregnancy, Mollie and Albert keep their indiscretion secret, under the idea she was artificially inseminated, and that Albert plans to leave his wife Beth and their two children to be with her. Mollie and her friend Rona happen to catch Albert fooling around with his interior decorator Melissa and he admits he is planning on living with her after his divorce is finalized. Mollie leaves upset, and immediately goes into labor. She gets into a cab where the driver, James Ubriacco, recklessly speeds through downtown traffic in order to get her to the hospital on time, and he is inadvertently a witness to her son Mikey's birth. Mikey then begins to make commentary on his life and interacts with things through an inner voice which can also communicate with other babies.

In present-day Antioch, former minister Travis Jordan still lives, after losing his faith in God three years before when his beloved wife was murdered and the criminals never found. Suddenly, miracles happen in the little town: the new veterinarian's son survives a van accident without a single scratch; Travis' dog Max revives after being buried; a paraplegic walks; a scarred teenager and her police officer father (brain tumor) heal, outside and inside. A group of young men in Africa, undergo a rite of passage symbolizing their change from boyhood into manhood. In rural China, a heavily pregnant Jin Ping is caught by Chinese military men with crates of black market blood in her van. The blood is destroyed and she is subsequently gang raped. Jin Ping is then shown visiting a village and convincing the inhabitants to give blood for $5 each. They all agree, except Tong Sam, a rice farmer, as he is unable to give blood as he is sick. However, as Jin Ping's equipment is not safe, most of the people in the village contract HIV and die of AIDS, including Tong Sam's family. The military men led by Xuan arrive in the village to help with the disease, and they help Tong Sam grow rice, which he then gives out to his remaining neighbors. Government officials arrive in the town to test people for AIDS, but the testing is $10 per person, which one neighbor thinks is a scam because it "cost $5 when she gave me the virus.

Prior to Christmas break, an unnamed female student (Emily Blunt) at a Pennsylvania university uses the campus ride share board to find a ride home to Wilmington, Delaware on 23 December. She finds a ride, which turns out to be a male student (Ashton Holmes), also unnamed ("the guy" in the credits), who is also driving to his home in Wilmington. His car is old and in poor condition, with the trunk full of his possessions (he tells her he is being kicked out of his apartment). It soon becomes apparent that she is somewhat arrogant and anti-social and his attempts to start a conversation go nowhere. It also becomes apparent that he knows quite a lot about her and possibly that he does not actually live in Wilmington as he claims. He tells her that they have a philosophy class together, although she has never noticed him.
