Raju and has music composed by R. P. Patnaik. The film was declared as Blockbuster Hit after Chitram and Nuvvu Nenu. It was a remake of the 1946 Bollywood film Anmol Ghadi, which was again remade in Hindi as Jeena Sirf Merre Liye. The film also lead to several other remakes.

Janaki (Reema Sen) and her sister are NRIs who want to join a PU college in AP. Janaki stays with her uncle and procures the admission in the same college as Ramana (Uday Kiran), a die-hard music fan. When Janaki first sees him in the music room practicing they get attracted to each other. When family members of Ramana are away, Janaki happens to come to Ramana's house wearing a saree. As she does not know how to wear saree, all she does is drape it around her body . Ramana offers to teach her how to wear a saree then accidentally he puts hand inside her saree skirt which makes them both feel shy and attracted. In the process they consummate their passion. After a few days Janaki informs Ramana that she is pregnant. Ramana, along with his friends, hires a nurse to perform abortion on Janaki. When Ramana asks Janaki to prepare for the abortion, she refuses to do so as she says she wants a company of a kid. Janaki tells him that her mother used to tell her that when she dies she will be reborn as Janaki's child. Janaki is then told by the college Principal to take rest and write the exam following year. When Ramana's parents talk to Janaki's uncle he blames Ramana.

The movie begins with Raghupathi (Nassar) being introduced as the eldest of his three brothers - the other two being Raghava (Tanikella Bharani) and Rajaram (Vajja Venkata Giridhar) and sister Swarajyam (Rajitha) and her husband (Chandramohan) with Chandu (Ram) as the only son of Raghava and sole inheritor of the property of 'R S Brothers'. Chandu plans the elopement of his cousin Swapna (Tamannaah) with her boyfriend (Navdeep) due to which he is ousted from the house and is isolated from his family by Raghupathi. However, Chandu tries to re-establish links with his family members, but to no avail. Meanwhile, on the last day of his college, Chandu and his friends meet their friend "Google" Gopi (Ravi Varma) who is depressed because his girlfriend is getting married to someone else. Chandu decides to stop the marriage and marry off her to Gopi. However, they kidnap Pooja (Genelia D'Souza) instead of Gopi's girlfriend due to a mistaken identity. After being chased by some hooligans - who are after Pooja - they escape with the exception of Chandu and Pooja who end up in forest. Chandu falls for Pooja who also reciprocates her feelings.

Ajay (Nagarjuna Akkineni) is an adman (director and actor) who has a definite idea about how his future wife should be. He gets married to a village belle Kamakshi (Simran Bagga), handpicked by his mom from her native town Kovvuru. After marriage, Kamakshi meets with a hairline fracture. The entire family of Kamakshi lands up at Ajay's house to take care of Kamakshi. She has a beautiful and greedy sister called Lahari (Reema Sen). During that period Suma (Suman Ranganathan), the regular model of Ajay meets with a fracture and Lahari replaces her as the model opposite Ajay. During that moment, Lahari gets lured by the modeling profession and then falls in love with Ajay. But Ajay does not have any feelings for her. Kamakshi gets well and her family returns to their village. Lahari tells Kamakshi about her desire to marry Ajay. Kamakshi gets mad and returns home to her husband. Meanwhile, Lahari attempts suicide. Kamakshi, being very close to Lahari, promises Lahari that she will get her married to Ajay. The rest of the film is about how Ajay manages to save his marriage without hurting Lahari and how he gets back Kamakshi.

Kiran (Akaash) and Aishwarya (Rekha Vedavyas) have been neighbours for 20 years in Hyderabad. They study in the same college and immensely hate each other. When their hatred continues, Rekha's parents shift to Ooty. There, Aishwarya finds an unopened greeting card in her room which is addressed to Deepika (Tanu Roy). Through her enquiry Rekha tries to know about deepika's whereabouts but her trials go in vain. Somehow she meets deepika's college mate who is her best friend those days! Ravi, played by Uday Kiran, is the son of a multimillionaire in Hyderabad. Vasundhara (Anita), is the daughter of a milk supplier. Ravi and Vasundhara both study in same college. The film starts with Vasundhara showing hatred towards Ravi, as he is a sportsman who is not good in his studies. Over a period of time, her hatred turns into love. Seenu (Ram) enjoys fights right from his childhood. He appreciates violence. He wants to be like Manikyam (Pradeep Rawat), who was a goonda, as everyone, including his teacher, is afraid of him. When all the boys wanted to become doctors, engineers, lawyers and other professionals, Seenu wants to become a goonda. The rest of the story is all about how she finally accepts Sanjay's love.
