The movie was directed by Michael Curtiz and produced by Arthur Schwartz, with Jack L. Warner as executive producer. The screenplay was written by Charles Hoffman, Leo Townsend and William Bowers.
The music score by Ray Heindorf and Max Steiner was nominated for an Academy Award. The film features several of the best-known Porter songs, including the title song, "Night and Day", "Begin the Beguine" and "My Heart Belongs to Daddy".
Alexis Smith plays Linda Lee Porter, Porter's wife of 35 years. Monty Woolley and Mary Martin appear as themselves, and the rest of the cast includes Jane Wyman, Eve Arden, Alan Hale, Dorothy Malone, Donald Woods, and Ginny Simms.
The film is a highly fictionalized and sanitized version of Cole Porter's life, leaving out amongst other things references to his homosexuality. A later film biography of Porter, the 2004 De-Lovely with Kevin Kline and Ashley Judd, dealt more frankly (though some said not enough) with his sexuality and includes a scene where Kline and Judd as Cole and Linda Lee watch Night and Day before its release and comment on its lack of realism and "happy ending".

In the early days of World War II, Cohan comes out of retirement to star as President Roosevelt in the Rodgers and Hart musical I'd Rather Be Right. On the first night, he is summoned to meet the President at the White House, who presents him with a Congressional Gold Medal (in fact, this happened several years previously). Cohan is overcome and chats with Roosevelt, recalling his early days on the stage. The film flashes back to his supposed birth on July 4, whilst his father is performing on the vaudeville stage.

As Jerry Golding (a young Korean War veteran) scales the heights of show business, he breaks the heart of his father, who had hoped that Jerry would instead follow in the footsteps of six consecutive generations of cantors in their family. The story deals with the joys and tribulations of a travelling minstrel troupe known as the Merry Meadow Minstrels. Al Jolson plays as a blackface endman while Lowell Sherman plays as the interlocutor. Hobart Bosworth plays as the owner of the show, while his daughter, played by Lois Moran, serves as Al Jolson's love interest in the picture. Sherman's character, however, is also in love with Moran's. The show is in a miserable state until Jolson entertains a sheriff and manages to convince him to invest in the show. The show becomes very successful thanks to this investment and Jolson is eventually able to visit his mother. Some time after he returns, he tells Moran that he loves her and this causes Sherman to become jealous. After a heated argument between Jolson and Sherman over Moran, a character played by Mitchell Lewis, who is upset because he was caught cheating at cards, puts real bullets in Jolson's stage gun. Since Jolson pretends to shoot Sherman in the minstrel show act, Lewis knows that this will result in Sherman's death and that Jolson will be blamed for the murder. After Sherman is shot, Jolson is arrested but manages to escape and take a freight train out of town. Eventually, Lewis confesses to the crime and Jolson is thereby proven to be innocent.

Elvira Kent (Janis Paige) and her husband Michael (Don DeFore) suspect each other of cheating. For their wedding anniversary, Elvira books an ocean cruise to Rio de Janeiro but her husband claims that unexpected business will prevent him from going. Seeing an opportunity, Elvira pretends to take the trip alone, but in fact sends singer Georgia Garrett (Doris Day), a woman she'd met at the travel agency, in her place and under her name. By secretly staying behind, Elvira hopes to find out if Michael is indeed sneaking around behind her back. Michael, however, is suspicious over Elvira's supposed willingness to go on the trip alone, and so hires private detective Peter Virgil (Jack Carson) to see if she is sneaking around behind his back.

The film opens in Los Angeles, where Doug Blake (Carson) is dumped as a manager by Gary Mitchell (Bowman). He goes to New York City to find a new singer to replace Gary on the Hour of Enchantment radio show. While in New York, he discovers Martha Gibson (Day) turning records in a jukebox factory. He takes her to Los Angeles and tries to introduce her to Felix Hofer (Sakall). His efforts lead to a series of communication failures.
