At age 34, Army Air Force pilot Major Robert Lee Scott Jr. (Dennis Morgan) is considered too old to fly in combat, but he is recruited and volunteers to fly in a secret bombing mission from the Philippines against Tokyo, the Japanese capital. When the mission is cancelled after his arrival in India because of the fall of the Philippines, Scott is promoted to Colonel and is then assigned to fly transport aircraft. He flies dangerous, unescorted missions over The Hump from Burma to China in order to supply aviation gasoline and other much-needed supplies to the three squadrons of the American Volunteer Group, the Flying Tigers.

Captain Jeff Dakin (George Montgomery) is shot down over Germany on a bombing raid as he sees his brother, Danny (Richard Graham) serving on the same aircraft, shot dead as he parachutes out of the stricken aircraft. Imprisoned in a camp, Dakin conspires with Alexandra "Alec" Zorich (Annabella), a beautiful Russian doctor, and Captain Paul Husnik (Kent Taylor), a Czech resistance leader, to mount an escape. They escape during an air raid and make their way towards safety, but the Czech is not who he seems.

In New York City, German agents arrange for Jane Graystone (Nancy Coleman) to take a taxi driven by one of their own men. The abduction goes awry when the taxi collides with another vehicle. Both driver and passenger are taken to the hospital in an ambulance attended by intern Dr. Mike Lewis (John Garfield). On the way, Jane regains consciousness and claims to have amnesia; she cannot remember who she is. The driver reports this to his superiors. Mike is excited, as this is his area of study, and persuades Dr. Murdoch (Roland Drew) to let him take the case.

In late 1941, Captain Rick Leland (Humphrey Bogart) is court-martialed and discharged from the U.S. Coast Artillery after he is caught stealing. Freighter owner and captain Mike Dillon (Dana Andrews) reluctantly smuggles Jewish immigrants into Palestine, making it very clear to the Jewish leader, David Vogel (Stephen McNally), he is only doing it for the money. Dillon is annoyed to learn that he will have to go ashore to get paid the $8000 he is owed. When a British patrol boat arrives sooner than expected, Dillon is forced to join the Jews in their flight for freedom. There are casualties on both sides before the illegal refugees get away, including one of Dillon's men.
