The Queens of Comedy is both a sequel and spin-off film of The Original Kings of Comedy both created and produced by Walter Latham. The film follows four black female stand-up comedians at Memphis's Orpheum Theatre.

Laura Hayes opens the show and serves as MC. She tells family stories about grandkids, her mom, and her sisters. Adele Givens urges her audience to celebrate their flaws; she comments on this crazy world, her 92-year-old grandmother, and the need to take care when naming a baby. Sommore, recently released from jail, talks about kids, men, marriage, and why moms give their eight-year-old daughters a hula-hoop. Lastly, Mo'Nique celebrates big women and contrasts Blacks and Whites. Mo'Nique tries to give big women hope, that it is ok to be a big woman, and it is ok to dislike skinny women.

The film also cuts away to footage of the Queens on the town having fun. For one night only, eight years later, the ladies returned to Mo'Nique for a comeback on The Mo'Nique Show which aired on October 29, 2009.

Nashawn Wade (Kevin Hart) has a horrible experience with a typical airline: his dog is classified as checked baggage instead of a carry-on, he eats a horrible airline meal, his buttocks gets stuck in the toilet during turbulence, and Nashawn's dog is sucked through a jet engine (after accidentally opening the cargo door). In response, he sues the airline and is awarded $100,000,000 by the jury. He decides to use the money to start his own airline, called N.W.A. (Nashawn Wade Airlines), whose acronym and logo are a pop culture reference to rap group N.W.A. The airline, specifically caters to African Americans and hip hop culture. The terminal at the airport is called the Malcolm X terminal, the plane is a heavily modified Boeing 747SP, customized with low-rider hydraulics, spinners, blended winglets to compensate for the drag from the decorations, and a dance club. The safety video is also a spoof of the Destiny's Child song "Survivor".

Diana Armstrong (LisaRaye) arrives at the scene of the raunchy, dysfunctional Players Club. The film has a very thin plot which is no more than a vehicle to link together the concert footage. It concerns the adventures of Ashley Wallace (Robert Hughes), a naïve DJ on Radio 2TW, who normally presents a through-the-night country and western-themed show. In spite of this, he is sent by the station's boss (Bruce Barry) to get an in-depth interview ("Not an interview, a dialogue", demands his boss) with the group, which is to be aired on the day ABBA leave Australia. Needless to say, Ashley, who has never done an interview before, singularly fails, mainly because he has forgotten to pack his press card, although the fact that he is unable to buy a ticket doesn't help matters either. Armed with his trusty reel-to-reel tape recorder, Ashley is forced to follow the group all over Australia, beginning in Sydney, and then travelling, in order, to Perth, Adelaide, and Melbourne, experiencing repeated run-ins with the group's very protective bodyguard (Tom Oliver), as well as his increasingly exasperated boss. Throughout the movie, we see Ashley interviewing members of the public, asking them if and why they like ABBA. Almost all the comments are positive, but he interviews a man who is driven mad by his ABBA-obsessed twelve-year-old, and another girl who thinks ABBA are over-the-top.
