Om Prakash Makhija (Shah Rukh Khan) is a junior artist in the 1970s Hindi film industry, in love with film star Shanti Priya (Deepika Padukone). One evening, Om views the premiere of Shanti's film Dream Girl, posing as Manoj Kumar. Later that night, Om becomes drunk and describes his vision of himself as a famous, wealthy film star winning the Filmfare Award for Best Actor to his friend Pappu (Shreyas Talpade) and some local children. At the shooting of a later film, Om rescues Shanti from a fire scene where of the fire has escaped control, and they become friends. One day, after following Shanti, he overhears her and producer Mukesh Mehra's (Arjun Rampal) conversation, that Mukesh and Shanti have married in secret and Shanti is pregnant and expects a traditional wedding. Mukesh pretends to be happy with the news, and invites Shanti at the set of their soon-to-be-made film Om Shanti Om. Mukesh promises to cancel the film, reveal their marriage to the public, and have a grand wedding on the set; but instead traps Shanti on the set and sets it on fire to prevent a financial loss and to protect his career. Shanti tearfully begs Mukesh to release her but he leaves anyway.

Set some six years after the events of Saturday Night Fever, former disco king Anthony "Tony" Manero is now 25. He has left Brooklyn, and is now living in Manhattan, staying in a flophouse while he works as a dance instructor and as a waiter at a dance club, looking for his big break in the modern dance productions on Broadway. The breakaway he has from his Brooklyn life, family, and friends seems to have matured Tony and refined his personality, specifically represented by his diminished Brooklyn accent and his avoidance of alcohol and profanity. However, certain attitudes have not changed, as with his most recent girlfriend, the forgiving Jackie (Cynthia Rhodes). In Sultanpur, a picturesque village in Pakistan, villagers have gathered to watch a game of cricket between India and Pakistan on television. Among them is a pregnant woman (Meher Vij) who, after giving birth, names her daughter Shahida after Shahid Afridi, the player who had won the game for Pakistan. On an afternoon, a six-year old Shahida falls off a cliff, but is saved by an overhanging tree. She loses her ability to speak from the accident. Her worried mother takes her to the shrine of Nizamuddin Auliya, a Sufi saint, in Delhi, on the belief that it would regain the latter's speech.

The film opens with two cousins – Heena (Asin Thottumkal) and Bobby (Jacqueline Fernandez), the two daughters of the Kapoor family who hate each other very much – start quarrelling over something relating to their duties of Animals' Law Authority. Mabel Stanley (Kristy McNichol) is an introverted teenage girl yearning for popularity in a seaside community in Australia. She attends a local pirate festival featuring a swordplay demonstration led by a young curly-haired instructor (Christopher Atkins), who then invites her for a ride on his boat. The film's plot revolves around Kanthaswamy (Vikram), who is an Income Tax officer in the CBI, and his friends, granting the wishes of the needy who write letters to a local Lord Muruga temple by disguising himself as an anthropomorphic rooster. During a CBI income tax raid, he discovers a stash of black money owned by Ponnusamy (Ashish Vidyarthi), a rich and corrupted entrepreneur. Ponnusamy pretends to be paralysed to escape the questioning. Angered by the news that her father is paralysed, Ponnusamy's daughter, Subbulakshmi (Shriya Saran) goes after Kanthaswamy to seek revenge by pretending to be in love with him. Kanthaswamy, knowing Subbulakshmi's real intentions plays along in the game. This forms a cat and mouse chase between the two. In a comical role, Thengakadai Thenappan (Vadivelu) is also sought after by a local police inspector who tries to solve the mystery of "Lord Muruga saving the people". The fate of Kanthaswamy's disguise leads into the climax of the story. The local police inspector was able to bring out the truth that a CBI is involved in the role behind the secret work of helping the needy people although he was not able to confirm it. Kanthaswamy finds the password of Subbulakshmi's family bank account, in which the black money is illegally saved. He tricks Subbulakshmi into revealing the password and answers for the security questions indirectly. Kanthaswamy finally transfers the money to his account which would be used for helping the poor and needy people. Ironically, in the end, Subbulakshmi's father was paralysed at the end knowing that the password which was used by him to access his black money was changed. After that Subbulakshmi and Kanthaswamy were married and were living in northern India. Kanthaswamy saw some letters tied to a tree, just like the ones at the Lord Muruga temple. He reads one and puts it into his pocket with a secret smile, which meant he will pursue being the anthropomorphic rooster again.

Randhir Bakshi et Dev Malik, pères respectifs de Kaali (Shah Rukh Khan) et Meera (Kajol) sont chefs de deux gangs adverses de la Mafia, ils sont ennemis depuis de nombreuses années. Meera réussi à lui faire avouer toutes les informations sur la cargaison d'or lors d'une de leur discussion, elle décide de reprendre l'or le jour de son anniversaire, jour qu'elle pense idéal pour faire ce cadeau à son père. Kaali est déçu et se sent trahi, il décide tout de même de la voir une dernière fois afin de lui dire deux mots, lors de cette rencontre, il va lui sauver la vie, ce qui va la faire tomber amoureuse de lui à son tour. Malgré quelques hésitations Kaali finit par pardonner Meera, et ils finissent par sortir ensemble. Meera veut faire un pas de plus dans leur relation et ils décident de convaincre leur pères pour leur mariage.
