The film combines clips from Tarkovsky's films with footage of Tarkovsky on the set of his last film The Sacrifice and on his deathbed, during the final stage of his battle with cancer. The film mostly relies on images, with only sparse commentary, and concentrates mainly on giving insight into Tarkovsky's work and philosophy and on exploring the intersections between his private life and his work. The film starts with a scene from Tarkovsky first film Ivan's Childhood and ends with a parallel scene from his last film The Sacrifice. It shows the reunion of Tarkovsky with his son Andrei Jr., who had been allowed to leave the Soviet Union only after Tarkovsky was diagnosed with terminal lung cancer. In Lost Soul Gregory looks at the filming of The Island of Dr. Moreau, specifically the period during which director Richard Stanley spent on the project. Stanley was brought on to the project early but was fired only a few days after principal photography began and was replaced by John Frankenheimer. Lars von Trier prétend que le court-métrage Det perfekte menneske de Jørgen Leth sorti en 1967 est son film préféré. Dans le but de lui rendre hommage, mais d'une manière perverse, à la fois manipulatrice et créatrice, il propose à l'auteur du film original, son compatriote Jørgen Leth, de détruire et de reconstruire autrement ce court-métrage. Il défie Leth d'en tourner cinq remakes. Pour chaque projet, il lui impose de nouvelles « obstructions », consignes, restrictions, interdictions plus ou moins arbitraires, l'obligeant à repenser l'histoire, l'esthétique et les personnages de son film. Il s'ensuit à chaque épreuve un feed back entre les deux hommes où Lars von Trier, dans une sorte de position de « directeur de conscience », à la limite de l'humiliation, semble prendre à cœur de déconstruire et détruire ce que Jørgen Leth a réalisé.
