The film, distributed by 20th Century Fox, is the sequel to 2003's X2 and the third installment in the X-Men film series. It was directed by Brett Ratner, written by Simon Kinberg and Zak Penn, and features an ensemble cast, including Hugh Jackman, Halle Berry, Patrick Stewart, Ian McKellen, Anna Paquin, Ellen Page and Famke Janssen. The film's script is loosely based on two X-Men comic book story arcs: "The Dark Phoenix Saga" by writer Chris Claremont and artist John Byrne, and "Gifted" by writer Joss Whedon and artist John Cassaday, with a plot that revolves around a "mutant cure" that causes serious repercussions among mutants and humans, and on the resurrection of Jean Grey.
Bryan Singer, who had directed the two previous films, X-Men (2000) and X2, decided to leave to work on Superman Returns, as he had not even defined the storyline for a third film. Matthew Vaughn, who was initially hired as the new director, left due to personal and professional issues, and was replaced with Ratner. Filming began in August 2005 with a budget of $210 million, and was consequently the most expensive film at the time of its release. It had extensive visual effects created by 11 different companies.
X-Men: The Last Stand was released on May 26, 2006, to commercial success. It grossed approximately $459 million worldwide, becoming the seventh-highest grossing film of 2006 and the highest grossing film in the series, until it was surpassed by X-Men: Days of Future Past in 2014. Critical reception was mixed, with the acting and the action scenes receiving positive notice, and criticism directed at the screenplay, overuse of characters and style.

In 1944 German-occupied Poland, 13-year-old Erik Lehnsherr is separated from his parents upon entering a concentration camp. While attempting to reach them he causes a set of metal gates to bend towards him, as though attracted by a magnetic force, before being knocked out by guards. Decades later, Senator Robert Kelly attempts to pass a "Mutant Registration Act" in Congress which would force mutants to publicly reveal their identities and abilities. Present are Lehnsherr, now known as Magneto, and the telepathic Professor Charles Xavier, who privately discuss their differing views on the relationship between humans and mutants.

In the future, robots known as Sentinels are exterminating mutants and their human allies. A band of mutants, including Kitty Pryde, Colossus, Iceman, Bishop, Warpath, Blink and Sunspot, evade the Sentinels due to Pryde's ability to send a person's consciousness to the past. Pryde's group convenes with Logan, Storm, Professor Charles Xavier, and Erik Lehnsherr at a monastery in China. Pryde sends Logan's consciousness 50 years back in time to 1973 to prevent Mystique from assassinating Dr. Bolivar Trask, creator of the Sentinels. Following the assassination, Mystique was captured, and her DNA was used by Trask's company to improve the Sentinels, whose ability to adapt to any mutant power makes them almost invincible. Xavier and Lehnsherr advise Logan to find both of their younger selves for help.

In 1845, James Howlett, a boy living in Canada, sees his father being killed by groundskeeper Thomas Logan. The trauma activates the boy's mutation: bone claws protrude from his knuckles, and he kills Thomas, who reveals that he is James's real father. James flees along with Thomas's son Victor Creed, who is thus James's half-brother. They spend the next century as soldiers, fighting in the American Civil War, both World Wars, and the Vietnam War. In Vietnam, Victor kills a senior officer. James defends Victor and the two are sentenced to execution by firing squad, which they survive. Artist and graphics designer Patience Phillips (Halle Berry) is a meek people pleaser whose only support is her best friend Sally (Alex Borstein). She works for a cosmetics company called Hedare Beauty, which is ready to ship a new skin cream called Beau-Line, that is able to reverse the effects of aging. However, as Patience visits the factory where it is being manufactured, she overhears a discussion between the scientist, Dr. Ivan Slavicky (Peter Wingfield), and Laurel Hedare (Sharon Stone), the wife of company owner George Hedare (Lambert Wilson); speaking about the dangerous side effects from continued use of the product. In Tibet, following the First World War, an American named Lamont Cranston (Alec Baldwin), succumbing to his darker instincts during the war, has set himself up as a brutal warlord and opium kingpin under the alias of Ying-Ko (Mandarin Chinese for "Dark Eagle"). He learns to "cloud men's minds," a form of mystical, psychic hypnosis that allows him to influence others' thoughts and bend their perceptions so he cannot be seen, except for his shadow (since light itself cannot be deceived), hence his new alias.
