Le docteur Mabuse est un personnage littéraire créé par l'écrivain luxembourgeois d'expression allemande Norbert Jacques (1880-1954).

Psychologue, hypnotiseur, criminel se dissimulant sous de multiples masques et personnalités, il est un des avatars des personnages de « génies du mal » présents dans la littérature populaire de l'époque, comme le docteur Fu Manchu créé par le Britannique Sax Rohmer, ou Fantômas créé par les Français Pierre Souvestre et Marcel Allain. Il faut probablement voir dans le succès du personnage non seulement le reflet d'une mode littéraire mais également l'expression d'une critique envers un monde en mutation, aussi bien d'un point de vue économique que des sciences humaines.

Mais ce sont surtout les trois adaptations cinématographiques qu'en fit Fritz Lang (cf. ci-dessous) qui rendirent le personnage célèbre, et le fixèrent dans l'imaginaire collectif, d'abord allemand puis international, au point d'apparaître dans une dizaine de films jusque dans les années 1970. Par ses côtés critiques et esthétiques expressionnistes, et leurs impacts sur l'histoire du cinéma, la démarche de Fritz Lang et de son épouse et scénariste Thea von Harbou dans Docteur Mabuse le joueur (1922) est à rapprocher du Cabinet du docteur Caligari de Robert Wiene (1920).

Inspector Lohmann's eagerly awaited fishing holiday is put on hold when he is called to investigate the murder of a man found in a railway tunnel. He was an Interpol courier carrying proof of the Chicago Syndicate's planned cooperation with a European criminal organisation that was in an attache case chained to, but now missing from his body. The Inspector gets word from Washington D.C. that a female representative of the Syndicate, Mrs. Pizarro is currently in Europe to meet up with the European criminals, and that the Federal Bureau of Investigation is sending Special Agent Joe Como to liaise with the German authorities. Lohmann visits a prison to have a meeting with one of the prisoners, Alberto Sandro, who was a member of the European criminal organisation in an unsuccessful bid to learn more about the upcoming events.

A reporter is killed in his car on his way to work. Inspector Kras gets a call from his informant Peter Cornelius, a blind fortune-teller, who had a vision of the crime but not the perpetrator. Meanwhile, Henry Travers, a rich American industrialist, checks into the Luxor Hotel, which has been outfitted by the Nazis during World War II to spy on people in every room. He becomes involved with Marian Menil who is being threatened by her evil clubfooted husband. Hieronymus B. Mistelzweig, purportedly a salesman, who is also a guest in the hotel always seems to be lurking about. These disparate characters eventually get together to solve what appears to be the re-emergence of the long-dead Dr.
