The film starts with Richland (Feore), who was sent to visit a government facility on a remote island. He follows Peniston (Esposito) into the facility. The head scientist Dr.Bishop (Reineke) shows them the projects he is working on, including a shark and dolphin hybrid with the ability to adapt to any environment, as well as a human and shark hybrid (the creature). It breaks free from its tank and kills Dr. Bishop. Richland orders Peniston to kill it, but Peniston instead traps the Creature in a containment unit and dumps it in the sea.

The film begins when a couple disappear while having a romantic night out on a yacht near the Pacific Northwest resort community, Grave's Point. The next day, local fisherman Whip Dalton (William Petersen) finds the empty lifeboat from the missing yacht and finds a large claw stuck into the boat. Whip sends the claw to a university to be analyzed and it ends up in the hands of marine biologist Dr. Herbert Talley (Ronald Guttman), who comes to Grave’s Point claiming it is from the tentacle of a Giant squid. The island harbor master Schuyler Graves (Charles Martin Smith) tries to hire Whip to kill the squid, but he refuses, so Graves sends out Lucas Coven (Larry Drake) instead. Coven succeeds in slaying the squid and the carcass is promptly sold by Graves to Sea Land Texas owner Osborne Manning (Denis Arndt) then unknown to the fishermen the sonar that isn't being watched detects another squid. It turns out the squid is much bigger than the one they killed and instead of following the boat or attacking, it resubmerges.

The film opens in 1968 at St. Anne's Academy, a California Roman Catholic school, where a young girl named Deloris Wilson is scolded by Sister Immaculata (Lois de Banzie) for wisecracking and disobedience. The setting then changes to the present day, where Deloris Van Cartier is a lounge singer in a 1960s-themed act called The Ronelles (a parody of The Ronettes), who sing at The Moonlite Lounge of the Nevada Club in Reno, Nevada, run by her boyfriend, the mobster Vince LaRocca. After Deloris walks in on Vince having his chauffeur Ernie executed for betrayal, Vince orders his two henchmen Joey and Willy to kill her as well. Deloris flees Vince's casino to the local police station where Lieutenant Eddie Souther suggests she testify against Vince if he can be arrested and tried, but for now, she should go into witness protection until the time comes.

The reluctant Bilal is to be the DJ. Kid is then involved in an altercation with school bully Stab (Paul Anthony) and his two brothers Pee-Wee and Zilla (Bowlegged Lou and B-Fine). When Kid comes home, he tries to convince his father, "Pop" (Robin Harris) to let him go to the party. At first Kid's father relents, but soon grounds Kid when a note from Kid's school informs him of the fight he was in. Rather than miss the party of the year, Kid sneaks out while his sleeping father is watching Dolemite - not realizing that his father woke up just as he closed the door. On his way to the party, Kid runs into Stab and his brothers, he jumps over a fence to get away, ending up looking in a window where a fat man is having rough sex with his lady, and when he is discovered, Kid jumps back over the fence, and the three punks are shot at. Kid then ducks into an Alpha Delta Sigma reunion nearby to get away from them. Crashing the reunion, Kid has the DJ (George Clinton) scratch and mix a few of his old doo wop records so that he can liven the party with a rap, until Stab and the others turn up again. When trying to get away from Stab, he winds up knocking an older man down before attempting to make a run for it. However, Kid and the bullies are caught by the neighborhood police, who humiliate the four teenagers in front of the reunion party attendees before letting them go.
