Firmly in the tradition of American Westerns, it follows the lives of the two orphaned brothers. The older one, Buddy, sees his father being shot. A humanoid alien (Aamir Khan) lands on Earth naked on a research mission in Rajasthan, but is stranded when the remote control of his spaceship is stolen, though he manages to get the thief's cassette recorder, a National Panasonic RQ-565D. On the same day in Bruges, Jaggu (Anushka Sharma) meets a man named Sarfaraz (Sushant Singh Rajput) and falls in love with him. Venkat Ramakrishnan (Srikanth), Sevarkodi Senthil (Jiiva) and Panchavan Parivendan aka Pari (Vijay) are three first year engineering students who share a room in the hostel of Ideal Engineering College (IEC), Chennai. Venkat studies engineering to pursue his father's wishes over his own wish – to become a wildlife photographer, while Senthil studies engineering to get his family out of poverty. Pari, on the other hand, studies engineering for his simple passion for machines and devices. He believes that one should follow excellence, not success, as success will take care of itself if excellence is followed. However, this approach to studies is sneered upon by the dean of the college, Professor Virumandi "Virus" Santhanam (Sathyaraj). Virus and the faculty favour Pari's classmate, an NRI student Srivatsan aka "Silencer" (Sathyan), who believes in mindless rote learning over understanding to reach his goals of corporate status. Pari constantly spars with Virus and Silencer due to their differences in their educational approaches.

Farhan Qureshi (R.Madhavan) and Raju Rastogi (Sharman Joshi) are students at the Imperial College of Engineering (ICE), a highly regarded Indian engineering college. Farhan's passion is wildlife photography, but he pursues an engineering degree to appease his father. Raju chooses engineering with hopes of improving his family's financial situation, but his lack of self-confidence results in poor grades. Their third roommate and friend, Ranchodas 'Rancho' Shamaldas Chanchad (Aamir Khan), has immense interest in robotics, and invents in his spare time. Rancho regularly faces the wrath of the college's director, Dr. Viru Sahastrabuddhe (Boman Irani), known as "Virus", when he gives creative and unorthodox answers in class. He lives by the philosophy that one should pursue excellence, as when it is achieved, success follows. Virus is shown as an extremely strict person—so strict that he caused his own son's suicide by putting him under excess pressure. The star student of their class is Chatur Ramalingam (Omi Vaidya) nicknamed "Silencer", known for his incredible techniques of memorization but more notorious for his silent but malodorous flatulence in addition to his arrogant attitude. Murli Prasad Sharma alias Munna Bhai (Sanjay Dutt), a gangster who is in love with the voice of Janhavi (Vidya Balan), a radio jockey, devises a plan to meet her when she announces a quiz contest featuring the life and beliefs of Mahatma Gandhi, to be aired on 2 October, the birthday of Gandhi. To prepare for the contest, Munna's sidekick Circuit (Arshad Warsi) kidnaps and bribes a group of professors to provide the answers for Munna. After winning the contest, Munna is granted an interview with Janhavi wherein he presents himself as a professor of history and a Gandhi specialist. Janhavi subsequently asks Munna to present a lecture on Gandhi to a community of senior citizens who live in her home, called the 'Second Innings House.' To prepare for this event, Munna engages in intense study at a Gandhi institute. For three days and nights (and without food or sleep), Munna reads about the life and ideologies of Gandhi.

Inayat Khan (Sanjay Dutt) is the Senior Superintendent of Police responsible for the security of Srinagar, Kashmir. One day, his young son Irfaan (Yogin Soni) meets with an accident and is taken to a hospital. Unfortunately, due to a fatwa instigated by the leader of a terrorist group forbidding doctors to treat policemen, the doctors refuse to treat Irfaan. Khan angrily threatens to kill the doctors if they don't treat his son, but they still refuse, saying they are willing to die rather than letting their families be killed by the terrorist group. As a result, Irfaan dies, and Khan personally swears to put an end to the threat of the terrorist group. After finding out that the group of terrorists is taking refuge in the village of Dalgate, Khan and his men manage to attack and kill the criminals. Unfortunately, a family is caught in the crossfire, and they are killed as well. A young boy named Altaaf is the only family member who survives the shooting. He is severely traumatized by seeing his parents and sister dying in front of his eyes and is haunted by the memory of the masked policeman who shot at his family: that police officer is Khan. Altaaf falls unconscious and is jailed by Khan's men, much to Khan's anger, who then berates his officers for placing Altaaf in jail. Khan's wife Neelima (Sonali Kulkarni), having just lost Irfaan and feeling sorry for Altaaf, attempts to persuade Khan to let them adopt the boy. Khan, despite his fear that Altaaf may one day discover the truth and get revenge for it, reluctantly agrees, feeling extremely remorseful for killing Altaaf's family.
