Dix ans après, Tallahassee, Columbus, Wichita et Little Rock font toujours équipe et luttent ensemble pour survivre face aux zombies, qui avec le temps ont évolué : parmi les zombies simples sont apparus les « Homer », en surpoids et simples d'esprit, les « Hawking », capables de stratégie, les « ninjas », rapides et silencieux, mais surtout les « T-800 », plus féroces et résistants aux balles. Le groupe trouve un temps refuge dans la Maison-Blanche. Mais des tensions apparaissent dans le groupe : Columbus et Wichita vivent leur petite vie de couple jusqu'à ce que Columbus demande la main de Wichita, et Tallahassee « couve » Little Rock comme un père, alors que cette dernière, devenue jeune adulte, a des envies d'ailleurs et d'être avec des gens de son âge. Une nuit, Wichita et Little Rock décident de partir à bord de la voiture blindée et surarmée de Tallahassee.

Po reunites with his biological father and travels with him to a secret sanctuary of pandas where, to his surprise, he does not fit in. L'histoire se déroule cinq ans après les événements de Sharknado 3. La tempête porteuse de requins tueurs est de retour et la cible des squales est à présent Las Vegas. Après avoir ravagé la ville, le Sharknado se déplace et frappe plusieurs états du centre des États-Unis, sans façade maritime, qui n'ont pas l'habitude des cyclones se formant au-dessus de la mer, mais plutôt des tornades continentales. En se déplaçant et frappant diverses installations (barrage Hoover, champ de pétrole, transformateur électrique, centrale nucléaire), la tornade change de nature en se chargeant de matières de plus en plus dangereuses : poussières, pierres, pétrole, feu, électricité à haute tension, et même radioactivité. Toute la famille Shepard, enfin réunie, va devoir unir ses forces pour en venir à bout.

Invité à Londres par le gouvernement britannique pour participer à une conférence de l'OTAN comme spécialiste en combat contre les requins, Fin est entraîné par Nova à explorer un temple souterrain situé sous Stonehenge. Des peintures rupestres leur apprennent que l'humanité a déjà été confrontée aux Sharknado à la préhistoire, mais les humains ont conçu un artefact (en forme d'aileron dorsal de requin) pour contrôler ces tornades. En s'en emparant, Fin et Nova déclenchent involontairement un Sharkado mondial qui frappe successivement les plus grandes villes du monde : Londres, Rio de Janeiro, Rome, Tokyo... Leur fils Gil ayant été aspiré par la tornade à Londres, Fin et sa femme April la poursuivent à travers le monde, pour tenter de sauver leur fils et empêcher la fin du monde.

Fin Shepard and his ex-wife April Wexler are flying to New York City to promote How to Survive a Sharknado and Other Unnatural Disasters, a book April has written about the Los Angeles sharknado. As the plane comes in for a landing through a storm, it is battered by airborne sharks, losing an engine. Sharks enter the plane, killing passengers and crew, including both pilots. While Fin successfully lands the plane, April's hand is bitten off by a shark while she attempts to shoot it with an air marshal's handgun.

After Flint Lockwood and his friends save the world from the food storm in the first film, super-inventor Chester V, the CEO of Live Corp, is tasked to clean the island. He relocates Flint, his friends, and the citizens of Swallow Falls to San Franjose, California. Unbeknownst to Flint, the FLDSMDFR survived the explosion and landed in the center of the island, and Chester is determined to find it. Chester invites Flint, his biggest fan, to work at Live Corp, where he meets Chester's assistant Barb, a talking orangutan with human intelligence. Six months later, Flint humiliates himself during a promotion ceremony when his invention, the "Celebrationator", explodes. Meanwhile, Chester is informed that his search-parties on the island have been attacked by monstrous cheeseburgers named Cheespiders (a combination of a Cheeseburger and a Spider) which are learning how to swim. Seemingly fearing the world's inevitable doom, Chester tasks Flint to find the FLDSMDFR and destroy it once and for all. Despite Chester's demands to keep the mission classified, Flint recruits his girlfriend, meteorologist Sam Sparks; her cameraman Manny; police officer Earl Devereaux; Steve, a monkey who communicates via a device on his chest; and "Chicken" Brent. Much to Flint's dismay, his father Tim joins the crew and they travel to Swallow Falls on his fishing boat.

In 1837, the Pirate Captain (Hugh Grant), inexpert in the ways of pirates, leads a close-knit, rag-tag group of amateur pirates who are trying to make a name for themselves on the high seas. To prove himself and his crew, the Pirate Captain enters the Pirate of the Year competition, with the winner being whoever can plunder the most. After several failed attempts to plunder mundane ships, the Pirate Captain talks with his best friend and first mate, the Pirate with a Scarf (Martin Freeman), about giving up and selling baby clothes as a living, but is convinced to try boarding the next ship they find, which happens to be the Beagle. They find no treasure but succeed in capturing Charles Darwin (David Tennant). Darwin recognizes the crew's pet Polly as the last living dodo, and implores the Pirate Captain to enter it into the Scientist of the Year competition at the Royal Society in London for a valuable prize. The Pirate Captain directs his ship to London. The Pirates disguise themselves as Girl Scouts, so that they don't get caught and executed, as Queen Victoria has a strong hatred of Pirates.

As a cub, Alex the Lion was called Alakay and was the son of Zuba, the alpha lion. Though Zuba tries to teach Alakay to be a hunter, the cub is more interested in dancing, and is captured by poachers when Makunga, Zuba's rival, challenges Zuba to a fight for the position of alpha lion. Alakay is forced into a crate, and taken away despite Zuba's attempts to rescue him. The crate falls into the ocean where it drifts to New York. There, Alakay is renamed Alex and sent to the Central Park Zoo where he grew up, meeting Marty the Zebra, Melman the Giraffe, and Gloria the Hippopotamus. Most of the events of the first film are then described through a series of news flashes.

In Rivet Town, Herb Copperbottom, a dishwasher at Gunk's Greasy Spoon diner, races through the streets, elated that he is going to be a father. He and his wife, Lydia, after 12 hours of "labor", construct the baby. He is named Rodney, and he becomes a young inventor who dreams of making the world a better place. Rodney idolizes Bigweld, a master inventor and owner of Bigweld Industries. During Rodney's adolescence, he invents a gadget, "Wonderbot", intended to help his father clean the dishes at the restaurant. When Herb's supervisor unexpectedly confronts them, Wonderbot breaks dishes, causing Herb to be put in debt and Rodney to be dismissed. Rodney takes his invention to Robot City to see Bigweld and get a job as an inventor at Bigweld Industries, so that he can help his father pay back his supervisor. His father encourages him and confides that he has always regretted not pursuing his dream of becoming a musician. Rodney arrives in Robot City and meets Fender, a ramshackle robot who takes souvenir photos and sells maps to the stars' homes. After a spectacular but harrowing ride on the crosstown express, Rodney arrives at the gate of Bigweld Industries.
