At work, Clark Griswold (Chevy Chase) has invented a long life food preservative, earning him a large bonus check. Clark announces to his family that he is taking them on vacation. Enthusiasm wanes, however, when Clark says they are headed to Las Vegas, Nevada. His wife, Ellen (Beverly D'Angelo) and teenage daughter, Audrey (Marisol Nichols) have their doubts, as Las Vegas is not known for its family-friendly atmosphere, while teenage son Rusty (Ethan Embry) appears to be more eager. Upon travelling to Vegas, they run into the "woman in the Ferrari" (Christie Brinkley) who appeared in the first film. Clark is the only one who sees her, but then notices that she now has a child.

In 1968, Jim Douglas is a down-on-his luck racing driver, reduced to competing in demolition derby races against drivers half his age. Jim lives in an old fire house overlooking San Francisco Bay with his friend and mechanic, Tennessee Steinmetz, a jolly Brooklynite who constantly extols the virtues of spiritual enlightenment, having spent time amongst Buddhist monks in Tibet, and builds "art" from car parts. After yet another race ends in a crash (and Tennessee turns his Edsel into a sculpture), Jim finds himself without a car and heads into town in search of some cheap wheels. He is enticed into an upmarket European car showroom after setting eyes on an attractive sales assistant, Carole Bennett. Jim witnesses the dealership's British owner, Peter Thorndyke, being unnecessarily abusive towards a white Volkswagen Beetle that rolls into the showroom, and defends the car's honor, much to Thorndyke's displeasure. The following morning Jim is shocked to find that the car is parked outside his house and that Thorndyke is pressing charges for grand theft. A heated argument between Jim and Thorndyke is settled when Carole persuades Thorndyke to drop the charges if Jim buys the car on a system of monthly payments.

After an incident where he used questionable police tactics, Jeff Powers (Lou Diamond Phillips) is placed on probation. Upon hearing of his probation, a friend from the force later invites Jeff to join the Special Investigation Section, an elite and highly secretive LAPD unit designed to track and shut down high profile criminals. Jeff discovers that the group is actually a group of rogue cops who actually function like an unofficially sanctioned death squad and are given wide latitude when it comes to dealing with criminals. Although their official mission is to surveil criminals and arrest them in the act of committing a crime, the squad often resorts to brutality and murder to dispatch the subjects they are supposed to arrest.

Sam Quint (Tommy Lee Jones) is a former thief hired by the FBI to steal a computer disk which contains incriminating evidence against The Lucky Dollar Corporation of Las Vegas. After stealing the disk, Quint is pursued by Marvin Ringer (Lee Ving), another former thief and acquaintance who works for the company. At the same time, a prototype vehicle called the Black Moon, which can reach speeds of 325 MPH and runs on tap water, is being tested in the desert by Earl Windom (Richard Jaeckel). Quint and Windom later cross paths at a gas station, where Quint hides the disk in the back bumper of the Black Moon. Windom is hauling the Black Moon to Los Angeles, and Quint, still being pursued by Ringer and his men, follows Windom and his team there.

Brilliant surgeon Daniel Jekyll (Mark Blankfield) dedicates himself to medical research, hoping to unlock man's animal nature and increase his capacity for self-healing. Chicago Examiner reporter Hildebrand "Hildy" Johnson (Jack Lemmon) has just quit his job in order to marry Peggy Grant (Susan Sarandon) and start a new career, when convict Earl Williams (Austin Pendleton) escapes from death row just prior to his execution.
