Four gifted street magicians—J. Daniel "Danny" Atlas, Merritt McKinney, Henley Reeves, and Jack Wilder—are brought together by an unknown benefactor and, one year later, are performing in Las Vegas as "The Four Horsemen," sponsored by insurance magnate Arthur Tressler. For the finale of their first show, they declare they will rob a bank, and invite audience member Étienne Forcier, an account holder at the Crédit Républicain de Paris, to do it. Forcier is apparently teleported to his bank in France, where he activates an air duct that vacuums up the money and showers it onto the Las Vegas crowd. Upon the discovery that the money really is missing from the bank vault, FBI agent Dylan Rhodes is called to investigate the theft and is partnered with Interpol agent Alma Dray. They arrest the Horsemen, but are forced to release them due to lack of evidence.

In Fall 1995 in Detroit, Jimmy "B-Rabbit" Smith Jr. (Eminem), a young and unhappy blue-collar worker from a poor family, struggles with various aspects of his life. He has moved back north of 8 Mile Road to the rundown trailer home in Warren, Michigan of his alcoholic mother Stephanie (Kim Basinger), his little sister Lily (Chloe Greenfield), and Stephanie's abusive live-in boyfriend Greg (Michael Shannon). Although encouraged by his friends, who hail him as a talented rap music artist, Jimmy worries about his potential as a musician, particularly due to his lack of confidence. This caused him to panic during a rap battle one night at a club called The Shelter, and leave the stage humiliated.

An alcoholic air marshal named Bill Marks boards a Boeing 767 non-stop flight from New York City to London. He sits next to passenger Jen Summers with whom he engages in friendly conversations. After take-off, he receives a text message on his secure phone stating that someone will die every 20 minutes unless $150 million is transferred to a specific bank account. Marks breaks protocol and consults with Jack Hammond, the other air marshal, who concludes that the threat is not valid. Marks however consults the help of Summers and flight attendant Nancy to monitor the security cameras while texting the mysterious person in order to locate him. However, Hammond is texted by the unknown person who says that he knows what is in Hammond's briefcase. As Marks confronts Hammond in the rear lavatory, the latter is revealed to be smuggling cocaine in a brief-case. Hammond attacks Marks, who responds in self-defense and breaks Hammond's neck. This incident happens exactly at the 20-minute mark.

Walter Mitty is a negative assets manager at Life magazine who daydreams of adventures and has a crush on a coworker named Cheryl. Mitty works with photojournalist Sean O'Connell, whose images are highly regarded. O'Connell has sent Mitty his latest negatives and a wallet as a gift in appreciation of Mitty's work. O'Connell believes negative #25 captures the "quintessence" of Life and that it should be used for the cover of the magazine's final print issue as it converts to online status. The negative is missing, however, and Walter is forced to stall for time with corporate transition manager Ted Hendricks, who is handling the downsizing. While viewing the other negatives outside Life's offices, Cheryl approaches Mitty and suggests that he think of the negatives as clues to Sean's location. They look at three of them, including one of a person's thumb with a unique ring on it, and another of a curved piece of wood. A third picture of a boat leads Mitty to determine that O'Connell is in Greenland. Mitty promptly flies there to find him.

Thomas "Tom" Popper (Jim Carrey) is a divorced real-estate entrepreneur whose father traveled around the world during his childhood. He learns that his father has died during an adventure to Antarctica and discovers that his father's will asks for his son to be given a souvenir from his last adventure. The next week, a crate containing a gentoo penguin (Captain) shows up at his doorstep. Eventually, five more penguins arrive. Popper intends to give them away but changes his mind when his children, Janie and Billy (Madeline Carroll and Maxwell Perry Cotton), think that the animals are Billy's birthday present. That night, Popper meets with New York zookeeper Nat Jones (Clark Gregg), who asks for the penguins. Popper tries to stall him by asking him to collect the penguins another time. Jones agrees but not before warning that the conditions in Popper's apartment are not good enough to raise penguins. Popper is forced to pay the board of his apartment to keep the penguins in his apartment and lies to his arrogant neighbor Kent (David Krumholtz) about there being no animals in his home.

Eddie Morra (Bradley Cooper), a struggling author suffering from writer's block, living in New York, is stressed by an approaching deadline. Later, Eddie happens to run into Vernon (Johnny Whitworth), the estranged brother of Eddie's ex-wife, Melissa (Anna Friel). Vernon, involved with a pharmaceutical company, gives Eddie a sample of a new "smart drug", NZT-48. After taking the pill, Eddie finds himself able to learn and analyze at a superhuman rate and recall memories from his distant past, with the only apparent side effect being a change in the color of Eddie's irises while on the drug—his eyes becoming an intense shade of electric blue. Under the influence, he cleans his messy apartment and writes ninety pages of his book. The next day, the effects having worn off, he seeks out Vernon in an attempt to get more. While Eddie is out running an errand, Vernon is murdered. Eddie returns, calls the police and then discovers Vernon's NZT stash just before they arrive, taking it for himself. After giving a statement at the precinct, Eddie returns home and begins ingesting the drug daily. With the help of the drug's amazing effects, Eddie spends a few weeks cleaning up his life—finishing his book, getting fit, and making friends with a group of young jet-setters, who take him on vacation to Europe, where he mingles with the rich. During all this, Eddie tests out his enhanced learning abilities; he becomes a proficient piano player in just three days, as well as becoming fluent in several languages.

A successful New York City attorney, Gavin Banek, is in a rush to file a power of appointment, which will prove a dead man signed his foundation over to Banek's law firm. He has a collision on FDR Drive with another car, belonging to an insurance salesman, Doyle Gipson, who is also in a rush to a hearing to try to gain custody of his children and to prevent his estranged wife from taking them to Oregon. Banek tries to brush Gipson off with a blank check thereby disobeying the law. After Gipson refuses to accept the check and voices his desire to "do the right thing", that is, filing a police report and insurance claim, Banek strands Gipson, telling him, "better luck next time". After arriving to the court late, Gipson learns that it proceeded without him and that it did not go in his favor.
