In an unnamed American city, soon-to-be-retiring Detective William R. Somerset (Freeman) is partnered with short-tempered-but-idealistic Detective David Mills (Pitt), who recently transferred to the department. Together, their first case leads them to believe that they are chasing a serial killer whose killings each reflect one of the seven deadly sins. First, they discover an obese man who was tied to a chair and forced to eat food, and eventually, his own vomit, until his stomach exploded (representing gluttony). Next, they investigate the fatal bloodletting of a rich attorney, from whom a pound of flesh has been extracted (representing greed). Two days later, fingerprints left at the crime scenes lead them to an apartment, where they find an emaciated man strapped to a bed. Though he initially appears to be dead, they find he has been kept alive and immobile by the killer for one year to that day; a drug dealer and child molester before his capture (representing sloth). Though unable to learn anything from the insensate man, the detectives agree the killer has planned these crimes for more than a year.

Recently divorced Meg Altman (Foster) and her 11-year-old daughter Sarah (Stewart) have just purchased a four-story brownstone on the Upper West Side of New York City. The house's previous owner, a reclusive millionaire, installed an isolated room used to protect the house's occupants from intruders. The "panic room" is protected by concrete and steel on all sides, a thick steel door, and an extensive security system with multiple surveillance cameras, a public announcement system, and a separate phone line. On the night the two move into the home, it is broken into by Junior (Leto), the previous owner's grandson; Burnham (Whitaker), an employee of the residence's security company; and Raoul (Yoakam), a ski mask-wearing gunman recruited by Junior. The three are after $3 million in bearer bonds, which are locked inside a floor safe in the panic room.

Silvia Broome (Nicole Kidman) is an interpreter working at the United Nations in New York City. She was raised in the Republic of Matobo, a fictional African country, but has dual citizenship. Two hundred years after the events of Alien 3, military scientists on the outer space vessel USM Auriga create a clone of Ellen Ripley (Sigourney Weaver) using DNA from blood samples taken before her death. They extract the embryo of an Alien queen that had been growing inside her at the time of her death, raise it, and collect its eggs for further use. The Ripley clone is kept alive for further study. As a result of her DNA being mixed with the Alien's DNA during the cloning process, she develops enhanced strength and reflexes, has acidic blood and a psychic link with the Aliens.

Gil Pender, a successful but creatively unfulfilled Hollywood screenwriter, and his fiancée, Inez, are in Paris, vacationing with Inez's wealthy, conservative parents. Gil is struggling to finish his first novel, centered on a man who works in a nostalgia shop. Inez dismisses his ambition as a romantic daydream and encourages him to stick with lucrative screenwriting. Gil is considering moving to Paris (which he notes, much to the dismay of his fiancée, is at its most beautiful in the rain). Inez is intent on living in Malibu. By chance, they are joined by Inez's friend Paul who is described as both pedantic and a pseudo-intellectual, and who speaks with great authority but questionable accuracy on the history and art of Paris. Paul contradicts a tour guide at the Rodin Museum, and insists that his knowledge of Rodin's relationships is more accurate than that of the tour guide. Inez admires him; Gil finds him insufferable.

Richard (Leonardo DiCaprio), a young American man with a love of world travel, arrives in Bangkok, Thailand in search of adventure. Dean Corso (Johnny Depp), a New York City rare book dealer, makes his living conning people into selling him valuable antique books for a low price, and then selling them for profit. He meets with wealthy book collector Boris Balkan (Frank Langella), who has recently acquired a copy of The Nine Gates of the Kingdom of Shadows by 17th-century author Aristide Torchia, one of only three copies known to exist. According to legend, the book is Torchia's adaptation of an older book written by the Devil himself and purportedly contains the means to ritualistically summon the Devil and, through him, acquire invincibility and immortality. Balkan believes two of the copies are forgeries. He hires Corso to authenticate all three copies, and acquire the legitimate copy by any means necessary.

In 1928, a globally famous illusionist, Wei Ling Soo, performs in front of a crowd in Berlin with his world-class magic act. As he walks off stage the film audience sees that he is actually a British man named Stanley (Colin Firth). He berates his employees and is generally curmudgeonly towards his well-wishers. In his dressing-room, he is greeted by old friend and fellow illusionist Howard Burkan (Simon McBurney). Howard enlists Stanley to go with him to the Côte d'Azur where a rich American family, the Catledges, has apparently been taken in by a clairvoyant and mystic, Sophie (Emma Stone). In fact, the son of the family, Brice (Hamish Linklater), is smitten with Sophie, and his sister Caroline (Erica Leerhsen) and brother-in-law George (Jeremy Shamos) are concerned Brice is considering proposing marriage. Howard says that he has been unable to uncover the secrets behind her tricks and he admits that the more he watched her the more he believed she really has supernatural powers. So he would like Stanley, who has debunked charlatan mystics in the past, to help him prove she is a fraud.
