FBI agent Malcolm Turner (Martin Lawrence) is elated to learn that his stepson, Trent Pierce (Brandon T. Jackson), has been accepted to attend Duke University in Durham, North Carolina. However, Trent is uninterested and instead wants Malcolm to sign a recording contract for him since he is underage. When Malcolm refuses, Trent's best friends encourage him to ambush Malcolm on the job in order to obtain the signature. Malcolm, in an attempt to capture Russian gang member Chirkoff (Tony Curran), uses an informant named Canetti to deliver a flash drive to the gang, while Trent attempts to ambush Malcolm on the job. Canetti (Max Casella) reveals that the flash drive is empty and a duplicate is hidden with a friend at the Georgia Girls School for the Arts.

Two former backup soul singers, Louis Hinds (Samuel L. Sheryl Hoover (Toni Collette) is an overworked mother of two living in Albuquerque, New Mexico. Malcolm Turner (Martin Lawrence) has been assigned a desk job in public relations as an FBI agent, since he wants to live with his new wife, Sherry Pierce (Nia Long), during her delivery for the couple's new baby boy. Meanwhile, an incident occurs in Orange County, California, where Malcolm's old friend, Doug Hudson (Kirk B.R. Woller), has been killed while he was going undercover. FBI agent Kevin Keneally (Zachary Levi) is doing surveillance on a former U.S. Army military intelligence specialist named Tom Fuller (Mark Moses), who has since retired and is working for a private corporation called National Agenda Software. The FBI has soon discovered that Tom is developing a computer worm which will create backdoors into the databases of all the branches of the U.S. government. Affected by his friend's death, Malcolm asks FBI chief, Crawford (Dan Lauria), to put him on the case, but Crawford refuses and tells him to stay away for safety analysis. By eavesdropping via the webcam, Malcolm finds out that the FBI is sending one of the agents to infiltrate Fuller's house as a nanny. Giving Sherry the pretext of attending a safety conference in Phoenix, Arizona, Malcolm leaves for Orange County and takes the "Big Momma" costume with him.

Filmmakers James Hanlon and the Naudet brothers were originally filming Tony Benetatos, a probationary firefighter of the New York City Fire Department assigned to the Engine 7/Ladder 1/Battalion 1 Firehouse on Duane Street in Lower Manhattan with the intention of making a film about the "probie's" first experience as a firefighter. On the morning of September 11, the firehouse, under the direction of Battalion Chief Joseph Pfeifer, was called out on a reported "odor of gas" at Church and Lispenard Streets. Jules rode with Pfeifer to investigate, while Gedeon stayed behind at the firehouse with the "probie.

The film begins in an illegal underground dog-fighting arena in Korea, where an FBI agent named John Maxwell (Paul Giamatti) has been identified. John is ordered to be killed by a Korean mob boss, but is eventually rescued by his undercover partner and master of disguise, Malcolm Turner (Martin Lawrence). Suddenly, a group of FBI agents storm around the arena. Meanwhile, a criminal named Lester Vesco (Terrence Howard), who was originally serving a life sentence in prison for murder and armed robbery, escapes from his cell by killing the doctor and stealing his car. The FBI assigns Malcolm and John to capture Lester by sending them to small-town Cartersville, Georgia to stake out the house of a fat, elderly African American woman named Hattie Mae Pierce (Ella Mitchell; whom her friends call "Big Momma"), the estranged Southern grandmother of Lester's ex-girlfriend, Sherry Pierce (Nia Long), who supposedly aided Lester in his original bank robbery by giving him the key to the vault. After Big Momma unexpectedly leaves town to help her ill friend within a couple of weeks, Malcolm and John sneak into her house to plant security cameras and tap the phones. Sherry later calls Big Momma's house and Malcolm disguises his voice as Big Momma in order to lure Sherry and possibly obtain a confession. The plan works, in which Malcolm and John work together on a Big Momma disguise costume before Sherry's arrival the next day.

Kelvin "Kelley" Morse and Jasper Arnold become involved in a car race and accidentally damage a restaurant owned by Samantha Cavanaugh's parents. Both are sentenced to perform community service by repairing the damage. Although Kelley comes from a wealthy family and Jasper's parents are working-class, they soon find themselves fighting over the same girl, Samantha. While Jasper and Samantha have been courting publicly for years, in secret, Kelley and Samantha begin to spend time together. They soon find that they have more in common than they imagined, and they fall in love. Eventually, Jasper learns of their interlude and doesn't like it. During a trip to Kelley's home in Boston he reveals to Samantha that his mother killed herself. Samantha brings Kelley into the house and they sleep together. In the morning, after Sam makes Kelley breakfast, Kelley's father arrives and informs him he must attend college early and give up his fling with Samantha. Upon returning to the small town, Samantha's parents soon learn that their daughter has osteosarcoma, stemming from a track/field injury, and only a few months to live. Samantha tells Kelley that she thinks everyone has their own heaven and it is made of a combination of all the things we loved in life. She says that his mother has Kelley with her in her heaven. When Kelley learns the awful truth, he must decide if he should obey his father's wishes and go to college or stay by the side of the first girl he's ever loved. In the end he returns to be with Samantha during her final months of life. At her funeral, Kelley recites a passage from a poem he and Sam loved. The film closes with a shot of Samantha running through a field in her version of heaven.
