The movie opens with Daphne Blake trying to pass her driver's test and get her license. She is in the Mystery Machine. Velma Dinkley tells her she can do it, yet despite the encouragements, Daphne crashes the van into a tree. Later she and Velma win a lottery that takes people into space. The two girls soon find themselves going into space along with Billionaire Sly Baron, Trained Astronaut Shannon Lucas, Retired Astronauts Zip Elvin and Colt Steelcase, Alien Hunting Specialist Ridley, Football player Uvininous "U-boat" Botango, News reporter Clark Sparkman (who instantly takes a fancy to Velma, much to her annoyance) and of course Fred Jones, Shaggy Rogers and Scooby-Doo. Fred allies himself with Zip and Colt, much to their dismay. Shaggy and Scooby are happy to see U-boat. Shannon tells Daphne that she scored the highest on the space test she and her friends took, much to Velma's surprise. This causes Velma to become bitter towards Daphne.

In the present day, Lex Luthor and his Legion of Doom try to use cryogenic rays from orbital satellites to expand the Earth's polar ice caps in the Arctic Ocean, causing a massive drop in sea levels and creating new islands they intend to rule. Superman and the Justice League confront the Legion, dividing their forces to fight the Legion and destroy the satellites. In the ensuing space battle, Robin purposely crashes his spacecraft into the satellite array. Refusing to accept defeat, Captain Cold overloads the remaining satellite, which falls back to Earth and entombs Luthor in an ice sheet shortly before the satellite explodes. The League is triumphant and believes Luthor to be dead.

Velma gets a call from Cuthbert Crawley, a lawyer for Velma's family who tells her that she's inherited her great uncle's property and castle in Transylvania. Velma turns the offer down, replying she wants nothing to do with her uncle, puzzling the gang. Mr. Crawley understands, revealing the curse over the estate. He says that if anyone gets too close to the Baron legacy, then what they love the most will be destroyed. As they head outside, Fred hears a noise from the Mystery Machine, then it explodes and a mysterious character with a mask appears. Crawley reveals that it's the ghost of the Baron, Velma's old ancestor. The remains of the machine explode again, with words saying stay away from Transylvania, but Fred declares that is where they're going to find the person responsible for destroying the Mystery Machine, saying this time "it's personal".

Offering herself as a hostage, Lois Lane is caught in an aerial confrontation between her terrorist captors and the unpredictable Supergirl before Superman arrives to save the day. Soon after, knowing Superman's civilian identity, Lois attempts to get Clark Kent to make their relationship public despite his fear of the consequences, but their argument is halted by a Daily Planet staff meeting before Kent leaves after being alerted to an approaching meteor. Intercepting it, Superman learns the meteor is actually a robot that he promptly defeats before activating its beacon and taking it to the Fortress of Solitude. With help from a fearful Supergirl, Superman learns the robot is actually a drone controlled by a being named Brainiac, a Coluan scientist who subjected himself to extensive motor, skeletal and cybernetic enhancements, turning him from a human like, thin, and hairless being to a muscular, red eyed giant with computer like components and enhanced physical abilities compatible to Superman's. Supergirl, horrified at seeing Brainiac, reveals from her experience with the monster. Brainiac seized and miniaturized Krypton's capital city of Kandor prior to the planet's destruction with her father and mother attempting to track him down before they mysteriously lost contact with Krypton. She is now worried that Brainiac will do to the world what he did to Kandor.

Scooby and Shaggy take the gang to a comic book convention called the "Mega Mondo Pop! Kantmiss Evershot practices archery in the forest, but her boyfriend, Dale, surprises her; the arrow accidentally hits the Wizard of Oz. As they return to District 12, where the residents fight over food, kids who are afraid of getting picked for the Games are injuring themselves by smashing themselves with pans or cutting off their feet and still being content. At the Gathering for the 75th annual Starving Games, President Snowballs explains he separated America into districts to keep "stupid stuff" from happening again, such as Lady Gaga being president with her running mate as Nicki Minaj; due to this, the rich now dress in bright and odd clothing. He also reveals the winner will win an old ham which expired in 1979, a pickle he's taken a bite out of, and a coupon to Subway. During the drawing for District 12's contestants, trouble occurs as vulgar name puns get drawn first. Petunia, Kantmiss's sister, tricks Kantmiss into volunteering when she gets picked by crying crocodile tears; Dale tries volunteering too, but idiotic Peter Malarkey beats him to it. The night before the Games, the contestants are being interviewed by Stanley Ceaserman, the host of the Starving Games countdown show. A stylist named Cinnamon gives Kantmiss a red dress, but it turns out he mistook her for another contestant in the Games. When Peter reveals his affection for Marco, a contestant from District 1 who is favored to win, Kantmiss is so furious that she literally sticks a foot up his rear end. When Kantmiss appears on stage, a video reveals how Dale first fell in love with her. It reveals that he was attracted to the sight of her defecating in the woods, which the audience finds pleasant. When Kantmiss spins her dress and catches fire, she starts screaming and swearing. There are also sports commentators for the Games, Bob Hylon and Cleaver Williams, a past Games winner who got his nickname by using a meat cleaver to kill a contestant.

The Buddies, are on a school trip to Warwick's mansion with their owners, Alice, Billy, Pete, Sam and Bartleby. When the class go to the graveyard to see Pip's memorial stone, Billy finds himself scared of the appearance of the gravedigger, Mr. Johnston. Overhearing that the legend says that if you say 'Halloween Hound' three times in front of Warwick's portal/mirror, you would release the Halloween Hound, B-Dawg, who is a "scaredy cat" tries to overcome his fear by doing the legend. Pip the ghost puppy tries to warn the Buddies, but B-Dawg is terrified by Pip's ghost and accidentally completes the reciting. B-Dawg's owner, Billy hasn't come up with his Halloween projects and decides to do Warwick the Warlock. With the help of Sheriff Dan, Billy borrows Warwick's evidence file and staff. Billy's mum, Janice buys Billy his costume for trick or treating which is a Christmas Story-esque bunny suit, having had misheard him saying, "Hip Hop Rabbit," when he meant "Hip Hop Rapper." Billy goes as Warwick the Warlock and humiliates B-dawg in the bunny outfit. Billy meets up with his friends and the Buddies and they go trick or treating.
