The film series is a complicated serial with more twists than a maze that basically centers around a U.S. Air Force captain and his quest for missing microfilm that contains vital information. The heroic Buster Crabbe plays Captain Roger Drum, who shoots down an enemy plane on its way to Africa with the secret microfilm. Intent on revealing the subversive group that the message was for, Drum assumes the pilot's identity and flies to Africa himself and crashes in the jungle. He is rescued by the pacific Rock People, led by Princess Pha (Gloria Dea), and is renamed Thunda, King of the Congo, after he rings a temple gong in alarm. With the subversives believing Thunda is their missing pilot and under constant attack by another tribe called the Cave Men, our hero plots to bring down the subversives who are searching for a new metal more radioactive than uranium. During the siege of Richmond, Virginia, in the American Civil War, POW Capt. Cyrus Harding escapes from his Confederate captors in a rather unusual way – by hijacking an observation balloon. In his escape, Harding is accompanied by sailor Pencroft, his nephew Bert, writer Gideon, loyal soldier Neb, and a dog. A hurricane blows the balloon off course, and the group eventually crash-lands on a cliff-bound, volcanic, uncharted (and fictitious) island, located in the South Pacific, with very unusual inhabitants. They name it "Lincoln Island" in honour of American President Abraham Lincoln.

Roar of the Iron Horse had hero Jock Mahoney in his first leading role, playing Jim Grant, a railroad agent, opposing a typical hardfaced German, Karl Ulrich, called The Baron. Head of a strong ring in America, the infamous Baron was thwarted time and time again as he tried to sabotage the building of the transcontinental railroad with all the means to his scope, strategically bribing the local Indians into doing his dirty work. Mahoney was an ideal choice to play the lead character, and the role marked the beginning of a long and rewarding association with Columbia Pictures.

Lex Luthor, the Atom Man, invents a number of deadly devices to plague the city, including a disintegrating machine which can reduce people to their basic atoms and reassemble them in another place. But Superman manages to thwart each scheme. Since Kryptonite can rob Superman of his powers, Luthor decides to create a synthetic Kryptonite and putters about obtaining the necessary ingredients: plutonium, radium and the undefined 'etc.' Luthor places the Kryptonite at the launching of a ship, with Superman in attendance. He is exposed to the Kryptonite and passes out. Superman is taken off in an ambulance driven by Luthor's henchmen, and he is now under the control of Luthor. Superman is placed in a device, a lever is pulled, and the Man of Steel vanishes into "The Empty Doom" (which bears a similarity to the Phantom Zone of the comic books). Most of chapter 7 is a repeat of the origin story from chapter 1 of Columbia's first "Superman" serial, and this serial also finds a way to work in stock footage from Ken Maynard's 1936 Avenging Waters (also directed by Spencer Gordon Bennet). The serial features several elements re-used in later Superman features: the Man of Steel is exposed to synthetic Kryptonite during a public function, as he is in the motion picture Superman III. When he escapes from "The Empty Doom", the headline of the Daily Planet proclaims: Superman Returns, the title of the 2006 Superman movie.
