Purefoy has a son, Joseph (b. 1997) from his first marriage to the actress Holly Aird. With the documentary director/producer Jessica Adams he has a daughter, Rose, who was born in New York in 2012. They were married in Somerset, England in July 2014.He is a supporter of Yeovil Town.

The film opens in North Africa, 1600, with the English mercenary Solomon Kane as he leads his ship's crew into battle against the Ottoman occupiers of a fortress town. After defeating the defenders, Kane and his men raid the fortress, where most of the crew are killed by demons. Kane fights his way to the throne room, but, before he can loot the riches, he is confronted by a demon that tells him his soul is forfeit to Satan. Solomon rejects his fate and jumps out a window. Following this encounter, Solomon returns to England and finds sanctuary in a monastery. After a prophetic dream, the abbot apologetically expels Kane, and Kane travels by foot to his ancestral estate, from which he had been expelled in his youth after defying his father. Along the way, he is ambushed by robbers who mock his vow of pacifism and leave him for dead. He is found and treated by the Crowthorns, a family of Puritans traveling west to the New World. When the Crowthorns are slaughtered by corrupted followers of the evil sorcerer Malachi, Kane renounces his vows and swears to avenge their deaths and rescue Meredith Crowthorn, who has been marked by a witch and kidnapped by the Masked Rider, Malachi's lieutenant.

Dr. Victoria Frankenstein, a scientist experimenting on stem cells and biotechnology, is working to advance medical science so her eight-year-old child, William, has a chance to receive organ transplants, and possibly a cure. This is the Universal Xenograft Project, overseen by Professor Andrew Waldman and assisted by her friend, Ed Gore. In the process of an organ-growing experiment, Victoria inserts William's blood into the procedure, and the stem cells begin to grow at a rapid rate. The insertion of William's blood is unknown until Ed discovers a tooth within the purpose built tank in which the stem cells are cultivating, and Andrew is then alerted. X-rays within the tank show that an organism is growing, and for the intention of scientific advancement, it is allowed to live. However, a lightning strike cuts the power within the building, and the creature, referred to as "the UX", escapes.

30 days after the contamination of The Hive seen in Resident Evil (2002), the Umbrella Corporation unwisely sends in a research team to re-open the complex and investigate the incident, since no one survived except Alice and Matt Addison, and as Alice was experimented on, Matt was put into a mysterious "Nemesis Program".
