On the surface, Richard (Humphrey Bogart) and Kathryn Mason (Rose Hobart) appear to be a happily married couple. But on their fifth wedding anniversary, Kathryn accuses Richard of having fallen in love with her younger sister, Evelyn Turner (Alexis Smith), who is visiting them. He does not deny it, but has resigned himself to leaving things as they are, since he is certain Kathryn would not give him a divorce. At a party celebrating the couple's anniversary hosted by family friend and psychologist Dr. Mark Hamilton (Sydney Greenstreet), Evelyn meets with Mark's handsome young colleague, Professor Norman Holdsworth (Charles Drake). On the way home, Kathryn suggests to Evelyn that their mother is lonely, so Evelyn decides to move home. Distracted by this unwelcome news, Richard crashes their car and suffers a broken leg. He then decides to take desperate action.

In 1940, German-born engineer Kurt Muller (Paul Lukas), his American wife Sara (Bette Davis), and their children Joshua (Donald Buka), Babette (Janis Wilson), and Bodo (Eric Roberts) cross the Mexican border into the United States to visit Sara's brother David Farrelly (Donald Woods) and their mother Fanny (Lucile Watson) in Washington, D.C. For the past seventeen years, the Muller family has lived in Europe, where Kurt responded to the rise of Nazism by engaging in anti-Fascist activities. An absent minded Professor Downie makes a call upon Drummond as he is making plans for his much-delayed wedding to fiancee Phyllis Claverling. The professor informs Drummond that a fortune was buried in one of the walled off storerooms underneath his estate, and that Downie was in possession of a book written in code that would lead them to discover the treasure. Unfortunately for the professor, someone else also wanted the riches and Drummond once again is dragged into the plot as the code book is stolen, Professor Downie is murdered, and Phyllis is kidnapped.

The story is based on two distinct early 19th-century themes, including the attempts by abolitionists (Cooper and Raft) to end the slave trade. Although the U.S. Constitution ended the importation of slaves in 1808, slaves were still being brought into the country under foreign flags. The abolition of slavery by Great Britain helped reduce legal trade in slaves by putting the British navy into action against slave traders, but even Britain had its supporters of the trade (here represented by Wilcoxon, as a British naval officer acting for the slave interests). The collision between Cooper and Wilcoxon is complicated by Wilcoxon's sister (Dee) falling in love with Cooper.

In a 1914 lower class district of Paris, content sewer worker named Chico (James Stewart) is criticized by his fellow citizens for being an atheist. One day, he witnesses a young woman (Simone Simon) being brutally beaten by her sister (Gale Sondergaard) for not entertaining a senior wealthy man in her sleazy bar. He sympathizes with the girl, Diane, and stops the fight. Later, Father Chevillon (Jean Hersholt), who is worried about Chico's lack of faith, offers him a deal: he will grant him a street washer job if he takes in Diane and opens his mind about believing in God. Chico agrees, and he prevents Diane's sister from turning her in to the police by claiming that she is his wife.
