Engineer Aron Ralston (James Franco) drives to Utah's Canyonlands National Park for a day of canyoneering. On foot, he befriends hikers Kristi (Kate Mara) and Megan (Amber Tamblyn), and shows them an underground pool. After parting ways with the hikers, Ralston enters Blue John Canyon through a slot canyon. He slips and falls, and a boulder falls and traps his arm against the wall. Failing to move the boulder, he calls for help, but no one is around. He begins recording a video diary on his camera and using the larger blade on his pocket multi-tool to attempt to chip away at the boulder. He also rations his water and food.

In 1996, Jamie Randall (Jake Gyllenhaal) is fired from a Pittsburgh electronics store for having sex with his manager's girlfriend. His wealthy brother Josh (Josh Gad) announces at the dinner table at their parents' (George Segal and Jill Clayburgh) house that he has found Jamie a job as a pharmaceutical sales representative. After attending a Pfizer training program where he has sex with the instructor (Kate Jennings Grant), Jamie goes to work for the company and tries to get doctors to prescribe Zoloft and Zithromax. He is rebuffed, much to the dismay of his regional manager, Bruce (Oliver Platt), who sees Jamie as his ticket to the "big leagues" of Chicago. Bruce says if Jamie can get Dr. Knight (Hank Azaria) to prescribe Zoloft instead of Prozac, other doctors will follow his lead. Jamie tries to get access to Dr. Knight by hitting on his female employees until, exasperated, Dr. Knight unethically permits him to observe him at work, during which time he accidentally sees a disrobing patient, Maggie Murdock (Anne Hathaway), who suffers from early onset Parkinson's disease.

The film opens with on-screen text stating: "A true story". It is August 1941, and Nazi Einsatzgruppen are sweeping through Eastern Europe, systematically killing Jews. While working an undercover prostitute sting operation in a nightclub to arrest a pimp named Neptune, Miami-Dade Police detectives James "Sonny" Crockett and Ricardo "Rico" Tubbs receive a frantic phone call from their former informant Alonzo Stevens (John Hawkes). Stevens reveals that he's leaving town, and, believing his wife Leonetta to be in immediate danger, asks Rico to check on her. Crockett learns that Stevens was working as an informant for the FBI but has been compromised. Crockett and Tubbs quickly contact FBI Special Agent in Charge John Fujima (Ciarán Hinds) and warn him about Stevens' safety. Tracking down Stevens through a vehicle transponder and aerial surveillance, Crockett and Tubbs stop him along I-95. Stevens reveals that a Colombian cartel had become aware that Russian undercovers (now dead) were working with the FBI, and had threatened to murder Leonetta via a C-4 necklace bomb if he didn't confess. Rico, learning of Leonetta's death by telephone call, tells Alonzo that he doesn't have to go home. Hearing this, the grief-stricken Stevens commits suicide by walking in front of an oncoming semi truck.
