A massive meteor shower destroys the orbiting Space Shuttle Atlantis and bombards a swath of land from America's East Coast from South Carolina through Finland. NASA discovers that a rogue asteroid the size of Texas passed through the asteroid belt and pushed forward a large amount of space debris. The asteroid will collide with Earth in 18 days, causing an extinction event that will even wipe out bacteria. NASA scientists, led by Dan Truman, plan to trigger a nuclear detonation 800 feet (240 m) inside the asteroid to split it in two, driving the pieces apart so both will fly past the Earth. NASA contacts Harry Stamper, considered the best deep-sea oil driller in the world, for assistance. Harry travels to NASA with his daughter Grace, to keep her away from her new boyfriend and one of Harry's drillers, A. J. Frost. Harry explains he will need his team, including A. J., to carry out the mission. They agree to help, but only after their list of unusual rewards and demands are met.

Four days after the events of the first film, detective James Carter is on vacation in Hong Kong, visiting his friend, Hong Kong Police Force Chief Inspector Lee, as he was sent along with Lee in saving the life of the Chinese counsel's daughter in Los Angeles. The fun is put on hold when a bomb explodes at the United States Consulate General, killing two undercover U.S. custom agents inside. Inspector Lee is assigned to the case, which becomes personal when it is discovered that it somehow involves Ricky Tan, his late police officer father's former partner. Tan, who was suspected of having a role in Lee's father's death - though the connection was never proven - is now a leader of the Triads. In 1973, Sam "Ace" Rothstein (Robert De Niro) is a sports handicapper and Mafia associate who is sent to Las Vegas to run the Teamsters Union-funded Tangiers Casino on behalf of the Chicago Outfit. He hires old friend Billy Sherbert (Don Rickles) as his manager. In between, Ace and his friend, mob enforcer and caporegime Nicholas "Nicky" Santoro (Joe Pesci), narrate how the mob bosses control the Teamsters Union, which gives out money for casinos that they own, such as The Tangiers, and how they also drive off rival crews and get rid of cheaters. Ace becomes the Tangiers' de facto boss by taking advantage of lax gaming laws allowing him to work at the casino while his gaming license is still pending. He doubles the casino's profits, which are skimmed by the Mob before the records are reported to income tax agencies. The bosses are impressed with Ace's work and send Nicky to protect Ace and the whole business, along with Nicky's brother Dominick, Nicky's friend and subordinate Frank Marino (Frank Vincent), and the rest of Nicky's soldiers in his crew. Nicky, however, becomes more of a liability than an asset; his criminal activities- which he makes nearly no effort to conceal- and his violent and vicious temper quickly gets him banned by the gaming board from every casino, and his name is placed in the Black Book. In retaliation, Nicky gathers his own crew, opens a jewelry store and restaurant, begins running unsanctioned shakedowns and burglaries, and soon after is considered the mob boss of Vegas.

After the mysterious failure of the Discovery One mission to Jupiter in 2001, which resulted in the deaths of four astronauts and the disappearance of David Bowman, the fiasco was blamed on Dr. Heywood Floyd, who resigned his position as head of the National Council for Astronautics. While an international dispute causes tensions between the United States and the Soviet Union, both nations prepare space missions to determine what happened to the Discovery. Although the Soviet ship, the Leonov, will be ready before the American spacecraft Discovery Two, the Soviets need American astronauts to help board the Discovery and investigate the malfunction of the ship's sentient computer, HAL 9000, which caused the disaster. The US government agrees to a joint mission when it is determined that Discovery will crash into Jupiter's moon Io before Discovery Two is ready. Floyd, along with Discovery designer Walter Curnow and HAL 9000's creator Dr. Chandra, joins the Soviet mission.
