Gaylord "Greg" Focker (Stiller) is preparing to celebrate his twins' fifth birthday party. Things seem to go awry when Greg's father-in-law Jack Byrnes (De Niro) visits. Recently, Jack has been diagnosed with a heart condition and become embittered by his daughter Debbie's divorce from her husband, Bob (whose marriage was the social event in the first film and how Jack and Greg met), for cheating on her with a nurse. Chuck Levine, a womanizing bachelor, and Larry Valentine, a widower struggling to raise his two children, are two veteran New York City fire fighters. In 1873, New Mexico Territory, an unnamed loner (Daniel Craig) wakes up in the desert being injured with no memory and with a strange metal object on his wrist. Ricky Bobby (Will Ferrell) is a NASCAR driver who wants to go fast. Eddie Morra (Bradley Cooper), a struggling author suffering from writer's block, living in New York, is stressed by an approaching deadline. His girlfriend Lindy (Abbie Cornish), frustrated with his lack of progress and financial dependence, breaks up with him. Under the influence, he cleans his messy apartment and writes ninety pages of his book. The next day, the effects having worn off, he seeks out Vernon in an attempt to get more. While Eddie is out running an errand, Vernon is murdered. Eddie returns, calls the police and then discovers Vernon's NZT stash just before they arrive, taking it for himself. After giving a statement at the precinct, Eddie returns home and begins ingesting the drug daily. With the help of the drug's amazing effects, Eddie spends a few weeks cleaning up his life—finishing his book, getting fit, and making friends with a group of young jet-setters, who take him on vacation to Europe, where he mingles with the rich. During all this, Eddie tests out his enhanced learning abilities; he becomes a proficient piano player in just three days, as well as becoming fluent in several languages.

South Boston teenager Jason Tripitikas (Michael Angarano) is a fan of martial arts films and he awakens from a dream of a battle between the Monkey King (Jet Li) and celestial soldiers in the clouds. He visits a pawn shop in Chinatown to buy Wuxia DVDs and discovers a golden staff. On his way home, Jason is harassed by some hooligans, whose leader Lupo attempts to use him to help them rob the shop-owner Hop. Hop tries to fight the thieves with the staff, but is shot and wounded by Lupo. He tells Jason to deliver the staff to its rightful owner and Jason flees with the staff. He is cornered on the rooftop by the hooligans and almost shot too, but he is pulled off the roof by the staff and falls backwards onto the asphalt.

In 1873, New Mexico Territory, an unnamed loner (Daniel Craig) wakes up in the desert being injured with no memory and with a strange metal object on his wrist. After killing three drifters who think he might be worth bounty money, he takes their clothes, weapons and a horse. He wanders into the small town of Absolution where the local preacher Meacham (Clancy Brown) treats his wound. After the stranger subdues Percy Dolarhyde (Paul Dano), a volatile drunk who has been terrorizing the town, Sheriff Taggart (Keith Carradine) recognizes the stranger as Jake Lonergan, a wanted outlaw, and attempts to arrest him. Jake beats up the posse sent to take him in and nearly escapes, but a mysterious woman named Ella Swenson (Olivia Wilde) knocks him out. Taggart prepares to transport both Jake and Percy to Santa Fe for trial.

Ricky Bobby (Will Ferrell) is a NASCAR driver who wants to go fast. While he was working on the pit crew of Dennit Racing driver Terry Cheveaux (Adam McKay), Ricky replaces him and ends up finishing in third place, despite starting mid-race in last place. Ricky quickly soars to the top of NASCAR by achieving his fame and fortune at Dennit Racing and meeting his future wife, Carly (Leslie Bibb). Ricky eventually persuades Dennit to field an additional car and arranges to have his best friend, Cal Naughton, Jr. (John C. Reilly), on his team. While he and Cal are on top of the racing world, Ricky challenges his flamboyant French Formula One archrival, Jean Girard (Sacha Barone Cohen), who outperforms him on the track and becomes Dennit Racing's next top driver. Eddie Morra (Bradley Cooper), a struggling author suffering from writer's block, living in New York, is stressed by an approaching deadline. His girlfriend Lindy (Abbie Cornish), frustrated with his lack of progress and financial dependence, breaks up with him. Later, Eddie happens to run into Vernon (Johnny Whitworth), the estranged brother of Eddie's ex-wife, Melissa (Anna Friel). Vernon, involved with a pharmaceutical company, gives Eddie a sample of a new "smart drug", NZT-48. After taking the pill, Eddie finds himself able to learn and analyze at a superhuman rate and recall memories from his distant past, with the only apparent side effect being a change in the color of Eddie's irises while on the drug—his eyes becoming an intense shade of electric blue. Under the influence, he cleans his messy apartment and writes ninety pages of his book. The next day, the effects having worn off, he seeks out Vernon in an attempt to get more. While Eddie is out running an errand, Vernon is murdered. Eddie returns, calls the police and then discovers Vernon's NZT stash just before they arrive, taking it for himself. After giving a statement at the precinct, Eddie returns home and begins ingesting the drug daily. With the help of the drug's amazing effects, Eddie spends a few weeks cleaning up his life—finishing his book, getting fit, and making friends with a group of young jet-setters, who take him on vacation to Europe, where he mingles with the rich. During all this, Eddie tests out his enhanced learning abilities; he becomes a proficient piano player in just three days, as well as becoming fluent in several languages.

Micky Ward (Mark Wahlberg) is an American welterweight boxer from Lowell, Massachusetts. Managed by his mother, Alice Ward (Melissa Leo), and trained by his older half-brother, Dicky Eklund (Christian Bale), Micky has not had a particularly successful career: He's become a "stepping stone" for other boxers to defeat on their way up. South Boston teenager Jason Tripitikas (Michael Angarano) is a fan of martial arts films and he awakens from a dream of a battle between the Monkey King (Jet Li) and celestial soldiers in the clouds. He visits a pawn shop in Chinatown to buy Wuxia DVDs and discovers a golden staff. On his way home, Jason is harassed by some hooligans, whose leader Lupo attempts to use him to help them rob the shop-owner Hop. Hop tries to fight the thieves with the staff, but is shot and wounded by Lupo. He tells Jason to deliver the staff to its rightful owner and Jason flees with the staff. He is cornered on the rooftop by the hooligans and almost shot too, but he is pulled off the roof by the staff and falls backwards onto the asphalt.
