The film begins where its predecessor ended, with reigning don Shoaib Khan (Akshay Kumar) earning respect after assassinating his mentor, Sultan Mirza (Ajay Devgn). Ram (Girish Kumar) is a rich, city boy, born to billionaire parents and brought up in Australia. On the other hand, Sona (Shruti Hassan)is a traditional, simple desi girl from Punjab who is brought up by her only brother, Raghuveer (Sonu Sood). He is heartbroken when their father marries another woman and throws them out of the house, humiliating them on the way. Their mother dies and her tomb is built on the small land which they own until the zamindar tells them that it is his land, since their mother had taken a loan from the man. Widower farmer, Dayal Singh, based in Palheri in the District of Chandigarh, witnesses the exploitation of farmers at the hands of Zamindars. He decides to send his elder son, Aman, to the city to study and become a lawyer, while his second son, Jiggar, would live with him and assist him in farming. Years later, the district gets a visit from Sohan Seth, who wants to buy the farm and turn the area into a commercial zone. Aman returns home and is able to join hands with his father and brother in advising the locales against Sohan. Things get out of hand when Jiggar is arrested for Assault, and even though Aman defends him, is sentenced to five years in prison. After his discharge, Jiggar returns home to find his father bed-ridden, half of his body paralyzed, a number of farmers have committed suicide, while Aman, who has married his sweetheart-fellow lawyer, Priya, lives in the city, works for Sohan, and has been putting pressure on Dayal as well as other farmers to sell their respective farmlands.

Radhe (Salman Khan), a gangster with a mysterious past, murders on other's orders for money. He meets Jhanvi (Ayesha Takia) while she's doing fitness training and instantly falls in love with her. Even though their first meeting leads Jhanvi to think negatively about Radhe, she later begins to reciprocate his feelings. In 2006, eighteen-year-old Jamal Malik (Dev Patel), a former street child (child Ayush Mahesh Khedekar, adolescent Tanay Chheda) from the Juhu slum, is a contestant on the Indian version of Who Wants to Be a Millionaire? (see Kaun Banega Crorepati), and is one question away from the grand prize. However, before the ₹20 million (US$300 000) question, he is detained and tortured by the police, who suspect him of cheating because of the impossibility of a simple "slumdog" with very little education knowing all the answers. Jamal recounts, through flashbacks, the incidents in his life which provided him with each answer. These flashbacks tell the story of Jamal, his brother Salim (adult Madhur Mittal, adolescent Ashutosh Lobo Gajiwala, child Azharuddin Mohammed Ismail), and Latika (adult Freida Pinto, adolescent Tanvi Ganesh Lonkar, child Rubina Ali). In each flashback Jamal has a point to remember one person, or song, or different things that lead to the right answer of one of the questions. The row of questions does not correspond chronologically to Jamal's life, so the story switches between different periods (childhood, adolescence) of Jamal. Some questions do not refer to points of his life (cricket champion), but by witness he comes to the right answer.

Rahul Verma (Arjun Rampal) and Karan Srivastav (Zayed Khan) are business partners. The film begins with the discovery of dead body of Pooja Sharma (Amisha Patel), Rahul's wife. It is apparent that Pooja has committed suicide by hanging herself. However, before the postmortem, Pooja's body goes missing. As the investigation goes on, the film turns to a flashback, where it is revealed that Karan used to be Pooja's ex-boyfriend. Karan was brash and good looking, but he was extremely possessive of Pooja because in a fit of rage Karan stabbed a man with a fork and forced Pooja to leave her father (Alok Nath). This caused Pooja to break up with him.

On 9 March 1993 a small-time thug, Gul Mohammed, is detained at the Nav Pada police station and confesses to a conspiracy underway to bomb major locations around the city. The police dismiss his confession and, three days later, Bombay is torn apart by a series of explosions leaving 257 dead and close to 1,400 injured. Ajay (Sohail Khan) is head of his college gang named Aryans. A girl named Ayesha (Sameera Reddy) to which Ajay is introduced and become friends. Gradually, Ajay and Ayesha falls in love with each other. This enrages Ayesha's father (Kabir Bedi) because of his status. Ajay challenges to get Ayesha back to him in nick time. Mr.Verma's friend and business partner Chopra (Dalip Tahil) too gets enraged and decides to finish off Ajay, but Chopra has an ulterior motive, Chopra's son (Raja Bherwani) to get married by Ayesha only to get Verma family's wealth. So Chopra appoints a notorious criminal Bhaijaan (Sanjay Dutt) to finish off Ajay. But Bhaijaan refuses as he do not do any wrong deeds in month of Ramadan. So Bhaijaan appoints his two head henchmen Munna (Rajpal Yadav) & Chhote (Sarfaraz Khan). Ajay at first empowers them during a fight but no sooner the formers appoint a gang to beat up Ajay brutally and left him dead on streets. Soon Ajay recovers from his injuries and enters Verma's mansion. An enraged Ajay challenges Verma to get back Ayesha at any cost. Verma too keeps a condition, if within 24 hours he didn't get back to Ayesha, he will have to get out of her life forever. Ajay accepts this deal. Chopra again plans a conspiracy and arrests Ajay without any crime. The police inspector (Aashif Sheikh) first beats Ajay and the inspector gets equally beaten up brutally and Ajay runs from the jail. Now Chopra calls Chhote to kill Ajay and promises to give him loads of money but Chhote replies that Ramadan is ended and Bhaijaan will finish Ajay. This conversation gets taped by Ayesha's little sister. Ajay gets surrounded by henchmen and knocks each one but interrupted by Bhaijaan. Just before Bhaijaan is about to kill Ajay, the former says he is a lover and will die once. This made Bhaijaan save Ajay. The reason was Bhaijaan had a brother Irfan who committed suicide because Bhaijaan didn't care for his feelings. During Eid ul Fitr Bhaijaan thanks Allah for giving a boy like Ajay in turn of his deceased brother and decides to help him. The very day, Ajay arrives to the Verma's mansion within 24 hours and decides to stop the wedding of Raman and Ayesha. Bhaijaan, who has a change of heart and helps him but soon interrupted by Chhote. Chhote disobeys Bhaijaan and soon a fight ensues. Bhaijaan requests Ajay to go but he is beaten up badly. Ajay, interrupts wedding and points gun on Verma. Soon Ajay and Ayesha gets married. Just about Verma is about to kill Ajay, Mini plays the tape of the deal to her father and soon Chopra and Raman are kicked off. Bhaijaan kills all the henchmen and finally slits Munna and Chhote with a sword. The film ends on a happy note with Ayesha and Ajay are united and Bhaijaan reconciles with Ajay.
