The story takes place at East Great Falls High, ten years after the events of American Pie. The new protagonists of this movie are three brand new hapless virgins: Rob (Hall), Nathan (Horton), and Lube (Hardesty). One night Rob accidentally sets fire to the school library, and finds "the Bible" (or Book of Love), the creation of Mr. Levinstein (Eugene Levy). Unfortunately for them, the book is ruined. The book had incomplete advice and sends them on a helpless journey to lose their virginity. After trying and failing for most of the time, they pledge to restore the book, and to do this, they must find Levenstein and all the other people who wrote in the book, and start the restoration.

Willard (Norm Macdonald) is an overworked, underpaid chauffeur who works for a mean-spirited pie heiress named Mrs. Crock (Elaine Stritch), just as his father did before him. All Willard wants for Christmas is a new uniform, as the one he currently wears is the one his father was buried in, but Crock gives him a cheap pair of cuff links and a pie instead, while lavishing expensive gifts on her business partner Chip Oswald (Sherman Hemsley) and her prized dog Muffin. Finally fed up with being mistreated, Willard and his best friend, local chicken restaurant owner Rusty (Dave Chappelle), concoct a scheme to kidnap the dog and hold it for a $1,000,000 ransom. Muffin attacks him, leaving a great deal of destruction and Willard's blood at the scene, and the plan fails when the dog later escapes.

Darryl Witherspoon (Marlon Wayans) is a college economics student, who does not have the advantages of his wealthy nemesis, Scott Thorpe (David Spade), or his best friend Tim LaFlour (Matthew Lillard), who has got a hockey scholarship. Darryl is so broke he donates four pints of blood in one day (playing a different character each time) and four vials of sperm in one day. At the beginning of the movie Casper is on a train full of ghosts, with no idea of where he is and is completely unaware that he is a ghost himself. Instead of going to Ghost Central, where the train is headed, he is kicked off the train. Billy (Lance Robinson) defies his mother (Karin Wolfe) and father (Craig Sechler) and makes a secret trip from Long Island to Philadelphia to sneak his grandfather (Abe Vigoda) out of a nursing home and bring him home for the holidays. The infamous vampire Count Dracula is expelled from his castle by the Communist government of Romania, which plans to convert the structure into a training facility for gymnasts (the head trainer declares that it will include Nadia Comăneci). The world-weary Count travels to New York City with his bug-eating manservant, Renfield, and establishes himself in a hotel, but only after a mix-up at the airport causes his coffin to be accidentally sent to be the centerpiece in a funeral at a black church in Harlem. While Dracula learns that America contains such wonders as blood banks, he also proceeds to suffer the general ego-crushing that comes from modern life in the Big Apple as he romantically pursues flaky fashion model Cindy Sondheim, whom he has admired from afar and believes to be the current reincarnation of his true love (an earlier being named Mina Harker).
