American burlesque performer Steve Martin (William Demarest) offers to play a song for his audience, if they agree to sing along. Only one person does sing, a young boy named Asa Yoelson (Scotty Beckett). Steve is bowled over by the boy's voice, but Asa realizes he should be singing at the synagogue with his father, Cantor Yoelson (Ludwig Donath). Asa arrives late, and is later reprimanded by his strict father. Asa is reluctant to explain where he was, but Steve Martin visits the Yoelsons' home. He explains that he heard Asa sing at the burlesque house, and that he wants Asa to be part of his act. Papa Yoelson refuses to consider it.

Spoiled heiress Ellen "Ellie" Andrews has eloped with pilot and fortune-hunter "King" Westley against the wishes of her extremely wealthy father, Alexander, who wants to have the marriage annulled as he is aware that Westley is only really interested in her money. Jumping ship in Florida, she runs away, boarding a bus to New York City, to reunite with her new spouse, when she meets fellow bus passenger Peter Warne, a freshly out-of-work newspaper reporter. Ellie agrees to the first choice. As they go through several adventures together, Ellie loses her initial disdain for him and begins to fall in love. When they have to hitchhike, Peter fails to draw attention until Ellie displays a shapely leg to Danker, the next driver. When they stop en route, Danker tries to steal their luggage; but Peter seizes his car. Nearing the end of their journey, Ellie confesses her love to Peter. When the owners of the motel in which they are staying notice that Peter's car is gone, they expel Ellie. Believing Peter has deserted her, Ellie telephones her father, who agrees to let her marry Westley. Meanwhile, Peter has obtained money from his editor to marry Ellie, but misses her on the road. Although Ellie has no desire to be with Westley, she believes Peter has betrayed her for the reward money, and agrees to have a second, formal wedding to Westley. On her wedding day, she finally reveals the whole story. When Peter comes to Ellie's home, Mr. Andrews offers him the reward money, but Peter insists on being paid only his expenses: a paltry $39.60. When Ellie's father presses him for an explanation of his odd behavior, Peter admits he loves Ellie, and storms out. Westley arrives for his wedding via Kellett K-3 Autogyro NC12691; but at the wedding ceremony, Mr. Andrews reveals Peter's refusal of the reward money to Ellie, sends her to Peter, and pays Westley off.

Infuriated at being told to write one final column after being laid off from her newspaper job, Ann Mitchell (Barbara Stanwyck) prints a letter from a fictional unemployed "John Doe" threatening suicide on Christmas Eve in protest of society's ills. When the letter causes a sensation among readers, and the paper's competition suspects a fraud and starts to investigate, editor Henry Connell (James Gleason) is persuaded to rehire Mitchell, who schemes to boost the newspaper's sales by exploiting the fictional John Doe. From a number of derelicts who show up at the paper claiming to have written the original letter, Mitchell and Connell hire John Willoughby (Gary Cooper), a former baseball player and tramp in need of money to repair his injured arm (by Bonesetter Brown), to play the role of John Doe. Mitchell starts to pen a series of articles in Doe's name, elaborating on the original letter's ideas of society's disregard for people in need.

During the Great Depression, Longfellow Deeds (Gary Cooper), the co-owner of a tallow works, part-time greeting card poet, and tuba-playing inhabitant of the (fictional) hamlet of Mandrake Falls, Vermont, inherits 20 million dollars from his late uncle, Martin Semple. The scheming attorney of his uncle, John Cedar (Douglass Dumbrille), locates Deeds and takes him to New York City. Cedar gives his cynical troubleshooter, ex-newspaperman Cornelius Cobb (Lionel Stander), the task of keeping reporters away from Deeds. Cobb is outfoxed, however, by star reporter Louise "Babe" Bennett (Jean Arthur), who appeals to Deeds' romantic fantasy of rescuing a damsel in distress by masquerading as a poor worker named Mary Dawson. She pretends to faint from exhaustion after "walking all day to find a job" and worms her way into his confidence.
