The parallel city of Monstropolis is inhabited by monsters and powered by the screams of children in the human world. At the factory of Monsters, Inc., employees called "scarers" venture into children's bedrooms to scare them and collect their screams, using closet doors as portals. This is considered a dangerous task because the monsters believe children are toxic and that touching them would be fatal. However, production is falling as children are becoming harder to scare and the company's chairman Henry J. Waternoose III is determined to find a solution. The top scarer is James P. "Sulley" Sullivan, who lives with his friend and assistant Mike Wazowski and has a rivalry with the ever-determined chameleon-like monster Randall Boggs. During an ordinary day's work on what is known as the "Scarefloor", another scarer accidentally brings a child's sock into the factory, causing the Child Detection Agency (CDA) to arrive and cleanse him. Former Army Ranger Cameron Poe is sentenced to prison for manslaughter for using excessive force on a drunk man while trying to protect his pregnant wife Tricia. Poe is paroled eight years later, and is to be released after being flown to Alabama on the Jailbird, a C-123K transport prison aircraft. Along with Poe are several other prisoners including his diabetic cellmate and friend Mike "Baby-O" O'Dell, who is being transferred (but not yet paroled) with Poe. The transfer is being overseen by U.S. Marshal Vince Larkin, as the transfer includes notorious criminal mastermind Cyrus "Cyrus The Virus" Grissom, gangster and Black Guerrilla member Nathan "Diamond Dog" Jones and mass murderer William "Billy Bedlam" Bedford for their transfer to a new Supermax prison. Chuck Levine, a womanizing bachelor, and Larry Valentine, a widower struggling to raise his two children, are two veteran New York City fire fighters. During a routine sweep of a burned building, a segment of floor collapses on Chuck. However, Larry quickly shields him from the falling debris, saving his life. Later, he and Chuck wake up in a hospital, where Chuck vows to repay Larry in any way possible. This incident serves as a catalyst for Larry's epiphany: death is both inevitable and unpredictable. This prompts him to significantly increase his life insurance policy, but he runs into difficulties naming his children as primary beneficiaries in his policy. He is told he should get remarried so his new spouse could be the primary beneficiary; however, it doesn't specify who he has to marry. Inspired by a newspaper article about domestic partnerships, Larry asks Chuck to enter a civil union with him. Although Chuck declines at first, he is reminded of his debt to Larry and finally agrees, entering a domestic partnership and becoming Larry's primary beneficiary in the event of his death. To their dismay, however, New York City investigators soon arrive to inquire about their abrupt partnership, under suspicion of fraud. Chuck and Larry decide to enlist the help of lawyer Alex McDonough, played by Jessica Biel, who suggests they get married and move in together to prove they're committed. Chuck reluctantly agrees. The pair soon travel to Canada for a quick marriage (so quick they use a nearby homeless person as the best man) and begin living together.

Maggie is the only cow left on the Dixon Ranch after Alameda Slim (a cattle rustler capable of stealing 500 in a single night) stole all the rest of Mr. Dixon's cattle. Dixon sells Maggie to Pearl, a kind and elderly woman who runs a small farm called Patch of Heaven. The local Sheriff arrives to tell Pearl that her bank is cracking down on debtors. Pearl has three days to pay the bank $750, or her farm will be sold to the highest bidder. Hearing this, Maggie convinces the other cows on the farm (Grace, a happy-go-lucky character, and Mrs. Caloway, who has had leadership gone to her head) to go to town to attempt winning prize money at a fair. While the cows are in town, a bounty hunter named Rico (who Buck, the Sheriff's horse, idolizes) drops a criminal off and collects the reward. Stating he needs a replacement horse to go after Alameda Slim while his own horse rests, he takes Buck. When Maggie find out that the reward for capturing Slim is of exactly $750, she convinces the other cows to try to capture him to save Patch of Heaven.

Later, Bones is driven out by Zee after he gets too fresh. When he leaves, he sees his kite in the doorway of Mr. Nebbercracker's house, but he and the kite are consumed, while retrieving it.

In 2065, Earth is infested by alien life forms known as Phantoms. Minneapolis car salesman Jerry Lundegaard (William H. Macy) is desperate for money. Later, he and Chuck wake up in a hospital, where Chuck vows to repay Larry in any way possible. This incident serves as a catalyst for Larry's epiphany: death is both inevitable and unpredictable. This prompts him to significantly increase his life insurance policy, but he runs into difficulties naming his children as primary beneficiaries in his policy. He is told he should get remarried so his new spouse could be the primary beneficiary; however, it doesn't specify who he has to marry. Inspired by a newspaper article about domestic partnerships, Larry asks Chuck to enter a civil union with him. Although Chuck declines at first, he is reminded of his debt to Larry and finally agrees, entering a domestic partnership and becoming Larry's primary beneficiary in the event of his death. To their dismay, however, New York City investigators soon arrive to inquire about their abrupt partnership, under suspicion of fraud. Chuck and Larry decide to enlist the help of lawyer Alex McDonough, played by Jessica Biel, who suggests they get married and move in together to prove they're committed. Chuck reluctantly agrees. The pair soon travel to Canada for a quick marriage (so quick they use a nearby homeless person as the best man) and begin living together.

Maggie is the only cow left on the Dixon Ranch after Alameda Slim (a cattle rustler capable of stealing 500 in a single night) stole all the rest of Mr. Dixon's cattle. Dixon sells Maggie to Pearl, a kind and elderly woman who runs a small farm called Patch of Heaven. The local Sheriff arrives to tell Pearl that her bank is cracking down on debtors. Pearl has three days to pay the bank $750, or her farm will be sold to the highest bidder. Hearing this, Maggie convinces the other cows on the farm (Grace, a happy-go-lucky character, and Mrs. Caloway, who has had leadership gone to her head) to go to town to attempt winning prize money at a fair. While the cows are in town, a bounty hunter named Rico (who Buck, the Sheriff's horse, idolizes) drops a criminal off and collects the reward. Stating he needs a replacement horse to go after Alameda Slim while his own horse rests, he takes Buck. When Maggie find out that the reward for capturing Slim is of exactly $750, she convinces the other cows to try to capture him to save Patch of Heaven.

Twelve-year-old DJ Walters spies on his elderly neighbor, Horace Nebbercracker, who confiscates any item landing in his yard. DJ's parents leave town for a dentist convention, leaving him in the care of a babysitter, Elizabeth "Zee". DJ's best friend Charles "Chowder" visits him, but accidentally loses his basketball on Nebbercracker's lawn. DJ is caught by Nebbercracker while recovering it, who rages at him before apparently suffering a stroke and being taken away by an ambulance. That night, DJ receives mysterious phone calls from Nebbercracker's house with no one on the other end. DJ eavesdrops on Zee's boyfriend Bones, who tells Zee about losing his kite on Nebbercracker's lawn when he was young and that Nebbercracker supposedly ate his wife Constance. Later, Bones is driven out by Zee after he gets too fresh. When he leaves, he sees his kite in the doorway of Mr. Nebbercracker's house, but he and the kite are consumed, while retrieving it.

In 2065, Earth is infested by alien life forms known as Phantoms. By physical contact Phantoms consume the Gaia spirit of living beings, killing them instantly, though a minor contact may only result in an infection. The surviving humans live in "barrier cities", areas protected by an energy shield that prevents Phantoms from entering, and are engaged in an ongoing struggle to free the planet. Minneapolis car salesman Jerry Lundegaard (William H. Macy) is desperate for money.
