Kabir Garewal (Arjun Rampal), a Casanova film maker and screenwriter is making multiple films (GUNS Trilogy) based on a thief's life and robberies. All his films have been highly successful. In order to shoot the third part of the trilogy, he goes to Malaysia, where he meets a London-based film maker Ayesha (Jacqueline Fernandez). Kabir and Ayesha get friendly and soon fall in love. When Ayesha finds out about Kabir's Casanova attitude, she breaks up with him and returns to London. Dejected, Kabir goes into depression and returns to Mumbai, leaving his film incomplete. After several attempts, Kabir is unable to find the perfect climax for his film. On his assistant Meera's (Shernaz Patel) suggestion, Kabir attends a film festival as part of the jury, where Ayesha's film is being screened. Ayesha thinks Kabir is following her and asks him to stay away from her life. After his father's (Anupam Kher) death followed by him being sued by his film's financiers, Kabir decides to move on and complete his film.

Ahana (Bipasha Basu) opens a hotel in Glendale, Himachal Pradesh. During the day of the opening party, she meets Kunal Anand (Imran Abbas), a novelist, and they become attracted to each other. When a couple and the hotel cook are killed by an unknown creature, Kunal decides to help Ahana. Romance culminates between Ahana and Kunal and they consummate their relationship. After a few peaceful days, the creature strikes again and wreaks havoc in the hotel; Ahana finds herself facing off against a ten-feet long creature with the legs and tail of a dinosaur, and the body and head of a human. Professor Sadanand (Mukul Dev) scares off the creature with a burning log.

The film opens by showing a large crowd waiting for Rahul Jaykar (Aditya Roy Kapoor) – a successful musician whose career is waning because of his alcohol addiction – to perform at a stage show in Goa. After nearly completing a song, he is unexpectedly interrupted by Aryan (Salil Acharya), who was losing his career due to Rahul's, during his performance. Rahul fights him, stops his performance, and drives to a local bar. He meets Aarohi (Shraddha Kapoor), a bar singer who idolizes Rahul. After noticing Aarohi looking at a photograph of Lata Mangeshkar in the bar, he assumes that she wants to become a singer. Impressed by her simplicity and voice, Rahul promises to transform her into a singing sensation, and asks her to never perform again in bars. Aarohi leaves her job and returns to Mumbai with Rahul, who convinces record producer Saigal (Mahesh Thakur) to meet her. When Aarohi calls Rahul, he is attacked and injured by some thugs, and is unable to receive her call. His friend and manager Vivek (Shaad Randhawa)says that news of Rahul's accident should not be leaked to the media, and instead publicises a false story that Rahul has left the country to participate in stage shows. When Aarohi attempts to contact Rahul again, Vivek ignores the calls. After two months of fruitlessly attempting to contact Rahul, a broken Aarohi is forced to sing in bars again because of her family problems.

The story carries on from the previous film— Inspector Chulbul Pandey (Salman Khan)— in charge of a local police station in Kanpur and resides there with his wife Rajjo (Sonakshi Sinha), brother Makkhi (Arbaaz Khan) and stepfather (Vinod Khanna). A boy gets kidnapped from a school therefore Chulbul deals with the problem. In Kanpur, an assassin murders a witness who is about to testify against a dreaded don. Sameer Behl (Shahid Kapoor) comes to Mumbai with Bollywood dreams, struggles through the day as a courier boy and keeps failing in auditions for advertisements. More stereotypes are stuffed in as he refuses to take help from his dad in Delhi, is thrown out from his rented flat by the landlord and comes away with his calculative companion. One day, a movie director, Rajeev Sharma, (Mohnish Behl) sees Sameer dancing and calls him to his office. Soon Sameer is signed as the male lead in his film and friend Tina (Genelia D'Souza) is selected as the choreographer on the same project. Homeless, Sameer sleeps in his car and works as a dance teacher in a school. Subsequently, Sameer loses his film and is disillusioned. Tina finds this out and asks him how he can spend his life like this. After some time, Tina tells him that she also quit the film and did not want to be the choreographer for that film because she saw him upset. Sameer realised the love Tina had for him. Tina encourages him to participate in a television talent-hunt show, the winner of which will win the same role that Sameer was to play. After some time he agrees to participate in it. After filling up the contest form, he also says "I love you" to Tina. In his screen-test, Sameer dedicates his monologue to his mother. After his monologue, during the interview, he comes to know that his father's shop in Delhi has been demolished. He promises Tina that he will come back for her and leaves. He leaves Mumbai to meet his father, heart-broken about the whole thing. After watching Sameer's performance in the TV, his father convinces him to go back to Mumbai and participate in the show.

The film opens with a suicide attempt by Assistant Commissioner of Police (ACP) Agnel Wilson (Randeep Hooda) on the pretext of the Bombay Bombings in 1993. When questioned by his superior over his actions, he breaks down and claims that the recent tragic events are his own fault. Wilson recounts that 18 years ago, when he was posted as the ACP in the Mumbai crime branch, his inability to take the necessary action led to the rise of Shoaib Khan (Emraan Hashmi) a dreaded gangster, who played a central role in the bombings. Throughout the film, Wilson narrates the story of 1970s Bombay, when it was ruled by a kind hearted smuggler Sultan Mirza (Ajay Devgn), and how Mirza's eventual downfall led to Shoaib's rise to power.
