Magobei Wakizaka is a samurai for the Sabai clan. A nearby island, Sado, boasts a rich gold mine which provides plentiful riches for the Tokugawa clan. When one of the gold ships sink, the local farmers recover some of the gold, intending to return it to the Tokugawa clan. However, Magobei's clan master, Rokugo Tatewaki, takes the gold and slaughters the farmers so they cannot report the gold stolen. Magobei is appalled. He promises not to report Rokugo to the shogunate in exchange for Rokugo 's promise to never do so again.

Gennosuke is a rebel samurai on the run, having fled his clan after assassinating a counselor. The daughter of the counselor, Misa, and her fiance, Daizaburo, pursue Gennosuke along with other samurai from Gennosuke's clan despite Gennosuke's obvious superiority as a warrior. A series of flashbacks reveals that Gennosuke was manipulated into committing the treason by one of the clan's higher-ranking samurai, who led Gennosuke to believe that the counselor's death would result in modern reforms to the clan and in Gennosuke's promotion to a full-fledged retainer, instead of a lowly foot soldier. In fact, the ranking samurai simply wanted the counselor killed so that he could succeed to the position himself. He had used Gennosuke to do the "dirty work", and then abandoned Gennosuke to face the consequences of the crime.

Yamamoto Kansuke (Toshiro Mifune) is a general of warlord Takeda Shingen (Yorozuya Kinnosuke), whose titular red banners are his trademark. In 1185, the Heike family fights against the Minamoto family. After a bloody naval battle in the Pacific Ocean, Yoshitsune Minamoto defeats the enemy and the survivors commit suicide. When the triumphant Yoshitsune arrives in Kyoto, his brother, the Shogun Yoritomo, is uneasy and orders his men to arrest Yoshitsune. However, Yoshitsune escapes with six loyal samurai led by Benkei and they head to the country of his only friend Hidehira Fujiwara. Near the border, after crossing the forest disguised as monks, their porter discovers that they are Yoshitsune and the six samurais and advises that the fearful Kajiwara and his soldiers are waiting for them in the border to arrest them. Yoshitsune disguises as a porter and at the barrier at the border, Benkei has to convince Kajiwara that they are six monks traveling to collect donation to build a large temple in Kyoto.
