'Don' begins, true to its title, as the story of one of the most powerful men in the business of crime. In spite of being on Interpol's Most Wanted list, Don remains elusive to the police. Besides the police, Don makes a few other enemies through his merciless approach to running his organization. Notably, Don kills one of his own men, Ramesh, when Ramesh decides to leave the business. This introduces Don to two new enemies, Kamini (Helen), Ramesh's fiancee, and Roma, (Zeenat Aman) Ramesh's sister. When Kamini seduces Don and attempts to have the police arrest him, her plan backfires as Don outsmarts her and the police in his escape. In the process, Kamini is killed. A shattered, revenge-seeking Roma gets her hair cut short, trains in judo and karate, then enters Don's gang after deceiving them into thinking that she too is on the wrong side of the law. Don is impressed with her fighting skills and allows her to work for him without suspecting any ulterior motive.

On a foggy night, Chandrashekhar (Navin Nischol), who is driving his car at high speed, meets with an accident. He walks to a nearby house to ask for assistance. When nobody answers the door, he enters the house and finds the dead body of a wheelchair-bound man. Standing nearby is a beautiful, young woman, Rani (Zeenat Aman), with a gun. She tells him that the dead man is her husband and she has murdered him. She asks Chandrashekhar to call the police. Intrigued, Chandrashekhar asks Rani to tell her what happened. She tells him that her husband, Thakur Ranjit Singh (Danny Denzongpa), was a cruel man who used to mistreat his family and used to shoot cats and dogs with his gun for amusement (which is why nobody in the house woke up upon hearing the gunshot). On this night, before Chandrashekhar arrived, they had an argument and when her husband threatened to shoot her, she tried to take the gun away from him. In the ensuing struggle, Ranjit got shot.

Ordered to stay away from Delhi's border for several years, two convicts, Kishan and Kanhaiya, decide to hitch a ride through a goods train to Bombay. While on this train, they come to the rescue of an elderly male with a female child, Munni. On his last breath, the elderly male asks them to take care of Munni as millions of rupees are involved. Greed overtakes both and they take the child with them in order to locate her next of kin. Unable to find anyone they advertise in the newspapers, and demand a hefty sum from Veer Singh, who answered the advertisement. What Kishan and Kanhaiya don't know that is that officially Munni is already dead, along with her mother, and brother, purportedly killed by their father, Ranvir Singh, when he suspected his wife's fidelity. Kishan and Kanhaiya find themselves immersed in a web of lies and deceit as they try to fathom what the truth really is. But before they could get to the bottom of this matter, their lives themselves are under threat - by a man who will stop at nothing to get what he wants.
