In 1795, Dr. Victor Frankenstein (Aden Young) creates a monster (Aaron Eckhart), a soulless creature patched together from corpses, and then rejects it, disgusted with what he's made. In a fit of rage, the creature kills Victor's wife Elizabeth (Virginie Le Brun) and Victor chases it to the Arctic to get revenge, but succumbs to the weather. The creature buries his creator in the Frankenstein family crypt and is then attacked by demons before being rescued by the gargoyles Ophir (Mahesh Jadu) and Keziah (Caitlin Stasey), who bring it before the gargoyle queen Leonore (Miranda Otto) and their commander Gideon (Jai Courtney). Leonore explains that they were created by the Archangel Michael to battle demons on Earth and protect humanity. They name the creature "Adam" and invite him to join them, but he declines and departs after being given heavy, baton-like weapons to protect him, as more demons will come after him. The weapons allow him to "descend" demons (destroying their bodies and trapping their souls in Hell) as they have the symbol of the Gargoyle Order carved on them.

Velma gets a call from Cuthbert Crawley, a lawyer for Velma's family who tells her that she's inherited her great uncle's property and castle in Transylvania. Velma turns the offer down, replying she wants nothing to do with her uncle, puzzling the gang. Mr. Crawley understands, revealing the curse over the estate. He says that if anyone gets too close to the Baron legacy, then what they love the most will be destroyed. As they head outside, Fred hears a noise from the Mystery Machine, then it explodes and a mysterious character with a mask appears. Crawley reveals that it's the ghost of the Baron, Velma's old ancestor. The remains of the machine explode again, with words saying stay away from Transylvania, but Fred declares that is where they're going to find the person responsible for destroying the Mystery Machine, saying this time "it's personal".

Lawrence Talbot (Lon Chaney, Jr.) is making an urgent call from London to a Florida railway station where Chick Young (Bud Abbott) and Wilbur Grey (Lou Costello) work as baggage clerks. Wilbur answers the phone and Talbot tries to impart to him the danger of a shipment due to arrive for the "McDougal House Of Horrors" (a local wax museum) which purportedly contains the actual bodies of Count Dracula (Béla Lugosi) and the Frankenstein Monster (Glenn Strange). However, before he is able to warn Wilbur, a full moon rises and Talbot transforms into a werewolf, who proceeds to destroy his apartment while Wilbur is on the line. Wilbur, thinking the call is just a prank, hangs up and continues on with his work day. Immediately thereafter, the actual Mr. McDougal (Frank Ferguson) shows up to claim the shipments and, fearing them damaged when Wilbur and Chick mishandle them, demands that the crates be delivered in person so his insurance agent can inspect them.

The Count, who introduces himself as "Baron Latos", explains that he has come to Visaria to find a cure for his vampirism. Dr. Edelmann agrees to help. Together with his assistants, Milizia (Martha O'Driscoll) and the hunchbacked Nina (Poni Adams), he has been working on a mysterious plant, the clavaria formosa, whose juices have the ability to reshape bone. Edelmann explains that he thinks vampirism can be cured by a series of blood transfusions. Dracula agrees to this, and Edelmann uses his own blood for the transfusions.

The film focuses on the exploits of the vengeful Dr. Gustav Niemann (Boris Karloff), who escapes from prison. He is helped by the hunchback Daniel (J. The residents of the village of Frankenstein feel they are under a curse and blame all their troubles on Frankenstein's monster. Rumors circulate about Ygor who is still alive and supposedly trying to revive the monster. The villagers pressure the Mayor into allowing them to destroy Frankenstein's castle. Ygor (Béla Lugosi) attempts to put up some resistance, but the villagers rush the gates and begin to destroy the castle. Ygor, fleeing through the catacombs, finds the monster released from his sulfuric tomb by the explosions. The exposure to the sulfur weakened the monster but also preserved him. Unseen by the villagers, Ygor and the monster flee the castle to the surrounding countryside; there they encounter a powerful thunderstorm. The monster is struck by a bolt of lightning, but instead of being harmed by it, he seems to be rejuvenated. Ygor decides to find Ludwig, the second son of the original Frankenstein, to help the monster.
