After the McManus brothers, Connor and Murphy (Sean Patrick Flanery and Norman Reedus), and their father, Noah (a.k.a. "Il Duce") (Billy Connolly), assassinated Joe Yakavetta, they fled to Ireland. Dale Denton (Seth Rogen) is a 25-year-old process server and habitual pot smoker. He makes a visit to the home of his drug dealer, Saul Silver (James Franco), to buy weed. Saul tells him that he may already know the identity of Dale's next service, Ted Jones (Gary Cole). Dale drives to Ted's house and witnesses Ted and a cop, Officer Carol Brazier (Rosie Perez), shoot an Asian man to death. Dale panics and flees the area, but leaves his roach at the scene which contains a rare strain of marijuana called Pineapple Express. Ted is able to identify the strain and sends his two henchmen, Budlofsky and Matheson (Kevin Corrigan and Craig Robinson) to dealer Red (Danny McBride), who tells them that he has only sold the pot to Saul.

Carl Taylor (Sheen) and James St. James (Estevez) are a pair of troublemaking garbagemen who dream of owning a surf shop. A Japanese minister of defence is traveling to Paris to sign a weapons contract between Japan and France, but first, he is visiting Marseille to view and rate the city police's anti-gang tactics (using fake attacks on him). During the visit, however, he is kidnapped by a group working for the Japanese yakuza. Emilien (Frédéric Diefenthal) is determined to rescue the minister and detective Petra (Emma Sjöberg), his girlfriend who was also kidnapped, and restore the honour of his department.
