Sixteen-year-old Minnesota high-schooler Juno MacGuff (Ellen Page) discovers she is pregnant with a child fathered by her friend and longtime admirer, Paulie Bleeker (Michael Cera). She initially considers an abortion. Going to a local clinic run by a women's group, she encounters outside a schoolmate who is holding a rather pathetic one-person pro-life vigil. Once inside, however, Juno is alienated by the clinic staff's authoritarian and bureaucratic attitudes. She is particularly offended by their calling her "sexually active", a term which she feels demeans the highly emotional event by which she became pregnant. All of this decides her against abortion, and she decides to give the baby up for adoption. With the help of her friend Leah (Olivia Thirlby), Juno searches the ads in the Pennysaver and finds a couple she feels will provide a suitable home. She tells her parents, Mac (J.K. Simmons) and stepmother, Bren (Allison Janney), who offer their support. With Mac, Juno meets the couple, Mark and Vanessa Loring (Jason Bateman and Jennifer Garner), in their expensive home and agrees to a closed adoption.

Berke Landers (Ben Foster) and his girlfriend Allison (Melissa Sagemiller) were the quintessential high-school couple who grew up together and eventually fell in love, but she breaks up with him immediately after the film begins. This leads to an opening musical number of "Love Will Keep Us Together" by Vitamin C, imagined by Berke. He seeks advice from his embarrassing parents Frank (Ed Begley Jr.) and Beverly Landers (Swoosie Kurtz), who are hosts of a relationship advice show called Love Matters, but they don't help with the situation and constantly focus on his sex life and sexuality throughout the film. Allison then starts a relationship with Striker (Shane West), a 'foreign' student who was once the lead singer of a boy band called "The Swingtown Lads". When Allison and Striker audition for the school's upcoming musical, Berke desperately tries to win Allison back by also auditioning for the play, despite having no theatrical talent and having a busy schedule as a member of the basketball team. Seventeen-year-old high school student Scott Howard (Michael J. Fox) is sick of being average and wishes he were special. His father, Harold (James Hampton), runs a local hardware store. Scott plays basketball for his high school's team, the Beavers, with a not-so-good win-loss record. The girl of his dreams, Pamela Wells (Lorie Griffin), is dating Mick McAllister (Mark Arnold), a jerk from an opposing high school team, the Dragons. After another of the team's losses, Scott begins to notice strange changes to his body. While at a party, Scott keeps undergoing changes and eventually he returns home, locks himself in the bathroom, and undergoes a complete change and becomes a werewolf, while his father demands that he open the door. He tries to refuse, only to finally give in and find his father has also transformed. Harold never told his son about the condition because "sometimes it skips a generation" and he was hoping it would not happen to Scott. Needing someone else to share his secret, Scott confides his best friend, Stiles Stilinski (Jerry Levine), who accepts Scott's werewolf status. However, Scott inadvertently reveals his transformation to the public at one of his basketball games. After shortly stunning the crowd with The Wolf, Scott goes on to amaze them with his newfound basketball skills.
