Following the events of Harold & Kumar Go to White Castle, Harold Lee and Kumar Patel set off on a flight to Amsterdam so that Harold can pursue a budding romance with his neighbor, Maria. Before they board their plane, they run into a friend from college named Colton, and Kumar's ex-girlfriend, Vanessa, who are getting married in Texas. During the flight, a woman suspects Kumar is a terrorist after mistaking his bong for a bomb. Sky marshals detain the duo and the plane diverts to Washington D.C. Ron Fox, an obsessive and racist Deputy Secretary of Homeland Security, sends them to Guantanamo Bay detention camp in Cuba, thinking they are agents of a joint Al-Qaeda and North Korean conspiracy, against the advice of NSA Vice-Chairman John Beecher. With the inadvertent assistance of two other prisoners, Harold and Kumar escape and board a Cuban refugee boat to Miami, Florida. They meet up with their college friend, Raza, who loans them a car so they can travel to Texas to get help from Colton, whose family has political connections.

Crime lord Oh Myung-gyu sends his subordinates, brothers Man-Seok and Jong-seok to retrieve the drugs; Jong-seok and his henchman Lum Rowan torture Hyo-jeong to find the drugs' location, then kidnap her and So-mi. Two gangsters, Du-chi and "Bear" go to Tae-sik pawn shop, but Tae-sik easily overpowers them. When he finds that Hyo-jeong and So-mi have been kidnapped, he gives them the bag, and Rowan kills Bear to set him up as the fall guy.

Ricky Verona (Jose Pablo Cantillo), an ambitious small-time criminal uses the opportunity to conspire with Carlito against Chelios; Verona will kill Chelios so the Triads do not retaliate, and then take Chelios's place as Carlito's new hired gun. While Chelios sleeps in his apartment, Verona, his brother Alex (Jay Xcala), and several henchmen break in and inject Chelios with a Chinese synthetic drug which inhibits the flow of adrenaline, slowing the heart and eventually killing the victim. Chelios wakes to find a recording left by Verona showing what he has done.

Tommy Chong plays Anthony "Man" Stoner, a jobless, marijuana-smoking drummer who is told to either get a job by sundown or be sent off to military school by his parents. Anthony leaves the house in a 1967 Volkswagen Beetle convertible (which had his father's Rolls Royce radiator grille on the front), a car which is subsequently left smoking on the side of the road. Anthony is picked up while hitchhiking by the equally enthusiastic smoker Pedro de Pacas (Cheech Marin). The license plate reads MUF DVR ("Muff Diver"). They share a large joint, which Man says is made with "mostly Maui wowie" and "Labrador" (essentially dog feces, as his dog, a Labrador Retriever, had eaten his stash). Police find their car parked on a traffic median with them in it, discover that they are clearly stoned and arrest them. At trial, the pair are released on a technicality after the judge's water pitcher is discovered to be containing vodka.

Suzanne is very reluctant to return to the woman she struggled to escape from for years after growing up in her shadow. The situation is not helped by the fact that Doris is loud, competitive, manipulative, self-absorbed and given to offering her daughter unsolicited advice with insinuating value judgments while treating her like a child.

Kumar receives a package with Harold's name on it at his apartment, and decides to deliver it to Harold. At Harold's house, they discover the item inside is a large marijuana joint.

On October 6, 1970, while on holiday in Istanbul, Turkey, American college student Billy Hayes straps 2 kg of hashish blocks to his chest. While attempting to board a plane back to the United States with his girlfriend, Billy is arrested by Turkish police on high alert due to fear of terrorist attacks. He is strip-searched, photographed and questioned. After a while, a shadowy American (who is never named, but is nicknamed "Tex" by Billy due to his thick Texan accent) arrives, takes Billy to a police station and translates for Billy for one of the detectives. On questioning Billy tells them that he bought the hashish from a taxicab driver, and offers to help the police track him down in exchange for his release. Billy goes with the police to a nearby market and points out the cab driver, but when they go to arrest the cabbie, it becomes apparent that the police have no intention of keeping their end of the deal with Billy. He sees an opportunity and makes a run for it, only to get cornered and recaptured by the mysterious American.
